一株极端嗜盐真菌的筛选及初步研究

摘要
利用含10%Na Cl的查氏培养基从晒干海鱼上分离到一株极端嗜盐真菌.为了对真菌进行理化鉴定,对分离菌株MF1和购买的标准菌株Phialosimplex salinarum DSM 27530在不同培养基上的培养特征、显微形态特征、耐盐性和在15%Na Cl的条件下的碳源利用能力进行分析检测,发现分离菌株生长盐度范围为0%33%,最适生长盐度为15%,并能够在15%Na Cl的条件下利用淀粉、脂肪、木质素、脱脂奶粉或纤维素为唯一碳源生长,说明该菌株在该条件下产生了相应的酶系.随后,对分离菌株和标准菌株的8个位点基因序列进行克隆与测序,最后进行多基因联合系统发育分析,确定分离菌株MF1属于Phialosimplex.由于分离菌株具有极高的盐度耐受能力和在高盐环境下产生丰富酶系的特性,因此该菌株在耐盐机理和工业应用上具有潜在的研究和应用价值.
        An extremely halophilic fungus was isolated from a dry marine fish using Czapek medium containing 10% Na Cl. In order to characterize the isolated fungi MF1,the morphological characteristics on different kinds of medium,micro-morphological characteristics,salinity tolerance abilities and substrate degration abilities under the condition of 15% Na Cl were discussed using the Phialosimplex salinarum DSM27530 as reference strain. The results showed that the isolated strain could grow at medium with 0% 33%Na Cl,optimum growth at 15%; and the strain was able to grow using starch,lipid,lignin,protein and cellulose as the sole carbon source under 15% Na Cl which indicating that the strain can produce the corresponding enzymes. Moreover,the multivariate phylogenetic analysis was carried out and the isolated strain was identified as a special of Phialosimplex. As the isolated strain has high salinity tolerance and the ability of producing active enzymes in high salt environment,it has potential value in the study of salt tolerance mechanism and application of industries.
