摘要
目的探讨丹参酮IIA(TanIIA)对膀胱癌T24细胞株体外增殖、凋亡以及细胞周期蛋白B1(CyclinB1)、半胱氨酸天冬氨酸蛋白酶-3信使核糖核酸(Caspase-3 mRNA)表达的影响。方法体外培养T24细胞,分别给予不同浓度(5 mg/L和10 mg/L)TanIIA,MTT法检测细胞增殖能力,AnnexinV/PI检测细胞凋亡,逆转录-聚合酶链式反应(RTPCR)检测细胞CyclinB1、Caspase-3 mRNA的表达。结果 MTT结果显示:5~10 mg/L TanIIA作用48 h均对T24细胞增殖有抑制作用,对细胞凋亡有诱导作用,而且CyclinB1mRNA下调及Caspase-3 mRNA上调,且均呈浓度依赖性。结论 TanIIA可能通过抑制CyclinB1mRNA和增加Caspase-3mRNA的表达水平,抑制膀胱癌T24细胞增殖及诱导凋亡,发挥抗膀胱癌的作用。
        【Objective】To explore the effect of tanshinone ⅡA(TanⅡA) on the proliferation and apoptosis of bladder carcinoma T24 cells and the expression of cyclin B1 and caspase-3 in vitro.【Methods】Bladder cancer T24 cells were cultured and treated with TanⅡA of different concentrations(5 mg/L and 10 mg/L) for 48 hours in vitro. TanⅡA-induced apoptosis was assessed by AnnexinⅡ/PI method and proliferation by MTT. The messenger RNA expression of cyclin B1 and caspase-3 in treated T24 cells and control were detected by real-time PCR.【Results】Treatment with TanⅡA resulted in a marked inhibition of proliferation and an obvious induction effect of apoptosis in treated T-24 cells on a dose-dependent manner. Significant down-regulated and up-regulated messenger RNA expression of cyclin B1 and caspase-3, respectively, in the treated T24 cells compared with the control were found on a dose-dependent manner.【Conclusion】The potential anti-tumor effect of TanⅡA was attested by proliferation inhibition and apoptosis induction of bladder carcinoma cells through reduction in the expression of cyclin B1 and increasing in caspase-3 in vitro.
