电针对脑卒中痉挛状态大鼠大脑皮质中谷氨酸与γ-氨基丁酸二者受体表达的影响

摘要
目的:观察和分析电针对脑卒中痉挛状态(SCA)大鼠大脑皮质中神经兴奋因子谷氨酸(Glu)与神经抑制因子γ-氨基丁酸(GABA)二者含量与表达的影响,从而论证针刺对SCA的缓解作用。方法:将32只SD大鼠随机分为空白组、假手术组、模型组、电针组(模型+电针法针刺阳陵泉、曲池),每组各8只。运用线栓法制作脑卒中痉挛状态模型,造模成功后第1天开始治疗,连续3d。以肌张力评分评价模型和针刺疗效,运用ELISA法分析大鼠大脑皮质中Glu、GABA的含量,RT-PCR法观察大鼠大脑皮质mGluR1a mRNA、GABABR1 mRNA的表达。结果:治疗前,模型组、电针组与空白组、假手术组比较,肌张力评分评分明显升高(P<0.01);治疗后,电针组与模型组比较评分明显降低(P<0.01)。模型组与空白组、假手术组比较Glu含量升高而GABA含量显著降低(P<0.01),电针组与模型组相比Glu含量明显降低而GABA含量显著升高(P<0.01,P<0.05)。模型组与空白组、假手术组比较大脑皮质mGluR1a mRNA的表达明显增强,而GABABR1 mRNA的表达明显减少(P<0.01),针刺组与模型组比较mGluR1a mRNA表达显著减弱,GABABR1 mRNA的表达显著增强(P<0.01)。结论:针刺可调节大脑皮质中Glu低表达和GABA的高表达,从而对SCA产生良好的治疗效果。
        Objective: To observe and analyze the effects of electro-acupuncture on the content and expression of neuronal excitatory factor Glu and neuro-inhibitory factor GABA in cerebral cortex of rats with SCA, so as to demonstrate the relieving effect of acupuncture on SCA. Methods: Thirty-two SD rats were randomly divided into control group, sham operation group, model group and electro-acupuncture group with model add electro-acupuncture at Yanglingquan(G34) and Quchi(LI11). There were eight rats in each group. The model of SCA was established by wire bolting method. Treatment started on the first day after successful modeling, for 3 days. The model and acupuncture efficacy were evaluated by muscle tone score, the content of Glu and GABA in cerebral cortex of rats with SCA were analyzed by ELISA, and the mRNA expression of mGluR1 and GABABR1 in the cerebral cortex of rats with SCA were observed by RT-PCR. Results: Compared with the control group and the sham operation group before the treatment, the muscle tone scores were significantly higher in the model group and electro-acupuncture group(P<0.01). After treatment, the score of electro-cupuncture group was significantly lower than model group(P<0.01). Compared with the control group and the sham operation group, Glu content increased and GABA content decreased in the model group(P<0.01), while compared with the model group the content of Glu was decreased and the GABA was increased in the electrocupuncture group(P<0.01, P<0.05). Compared with control group and sham operation group, the mRNA expression of mGluR1 in cerebral cortex was significantly increased in model group, while the mRNA expression of GABABR1 was significantly decreased(P<0.01). Compared with model group, mRNA expression of mGluR1 was decreased and mRNA expression of GABABR1 was increased(P<0.01). Conclusion: Electro-acupuncture could regulate low expression of Glu and high expression of GABA in the cerebral cortex, and thus had a good therapeutic effect on SCA.
