猪流行性腹泻毒株S基因分离鉴定及序列分析

摘要
根据NCBI上传的猪流行性腹泻病毒(porcine epidemic diarrhea virus,PEDV)[GenBank:JN547228(CH/S)],设计引物对阳性病料进行鉴定并回收,与pMD18-T构建重组载体并测序。对猪流行性腹泻病毒的基因多样性及序列保守性进行分析,一是以经典毒株为主,包括CV777、KPEDV-9和CH/S株等为代表的G1基因型,另一种是以近几年分离的毒株为主,包括HB3-CH2012、CXHZ-CH2013、HNCZ-CH2013等为代表的G2基因型。基因同源性分析比较结果表明,这4株PEDV之间的序列同源性为97.6%~100.0%,与经典株CV777的同源性为92.7%~92.8%。同源分析显示,PEDV不同毒株间的基因同源性仍高度相似,但PEDV流行毒株已呈现进化分支的趋势。
