摘要
目的优选阔叶十大功劳叶的小檗碱提取工艺。方法对阔叶十大功劳叶中小檗碱采用超声醇提,利用星点-响应面设计,以盐酸小檗碱含量为评价指标,选取料液比、乙醇浓度、超声时间3个因素进行中心组合设计,通过星点-响应面设计来筛选阔叶十大功劳叶中小檗碱的最优提取工艺。结果阔叶十大功劳叶中小檗碱最优工艺为:液料比13∶1;乙醇浓度54%;超声时间35min,提取2次,经3次验证试验,最佳工艺提取的功劳叶中小檗碱为0.683%(RSD=0.85%,n=3),与预测值0.691%比较,偏差为1.16%。结论星点设计-响应面法优化功劳叶中小檗碱的提取工艺,方法简单合理,且具有良好的预测性。
        Objective To optimize the extraction process of berberine from Mahonia bealei leaves by the central composite design-response surface method.Methods The ultrasonic ethanol extraction method was used to extract berberine from Mahonia bealei leaves.The central composite design-response surface design was used to make the central composite design with the three factors of solid-liquid ratio,ethanol concentration and ultrasonic time by taking the content of berberine hydrochloride as the evaluation index and screen the optimal extraction process of berberine from Mahonia bealei leaves.Results The optimal extraction process was as follows:the extraction process was conducted with the solid-liquid ratio of 13:1 and the ethanol concentration of 54% for 35 minutes of the ultrasonic time twice respectively,and the content of berberine in the optimal exaction process was0.683%(RSD=0.85%,n=3)with three validation tests,and the bias ratio was 1.16% when compared with the predicted value of 0.691%.Conclusion The extraction technology of berberine from Mahonia bealei leaves optimized by the central composite design-response surface method is simple and reliable with good predictability.
