摘要
目的:探究车前草不同药用部位抗炎、抗肿瘤及抗氧化作用活性差异,为进一步明确车前草不同药用部位的合理使用,扩大车前草临床用药,更好地开发利用车前草资源提供理论参考。方法:采用高效液相色谱法,以乙腈-0.2%甲酸水溶液为流动相进行梯度洗脱,建立车前根、车前叶、车前子及除去根、叶、子剩余部位的指纹图谱;采用脂多糖(LPS)诱导小鼠Raw 264.7细胞炎症模型,噻唑蓝(MTT)法测定细胞活力,Griess法测定细胞分泌一氧化氮(NO)的水平检测车前草不同药用部位抗炎活性;采用MTT法检测车前草不同药用部位抗肿瘤活性;采用1,1-二苯基-2-三硝基苯肼(DPPH)、2,2'-联氮-(3-乙基苯并噻唑-6-磺酸)二铵盐(ABTS)法检测车前草不同药用部位抗氧化活性。结果:车前草不同药用部位指纹图谱既有相似性,又存在一定差异性。药效学数据显示,车前草不同药用部位均具有较好的抗炎、抗肿瘤及抗氧化活性,其中抗炎和抗肿瘤活性车前叶活性最高,车前草剩余部位大于车前根,车前子活性相对较弱;抗氧化活性车前叶最高,车前根活性较低。结论:本研究明确了车前草不同药用部位醇提物指纹图谱及不同药用部位的抗炎、抗肿瘤及抗氧化活性差异,为进一步扩大车前草药材临床用药以及针对不同疾病合理利用不同药用部位精准用药研究提供一定的实验依据,为车前草药用资源的高效合理利用及药用新资源的开发提供理论依据。
        Objective: To explore the anti-inflammatory, anti-tumor and anti-oxidant activities of Plantaginis herba, in order to further clarify the rational use of different medicinal parts of the Plantaginis herba, expand clinical medication and provide a the reference for better development and utilization the Plantaginis herba resources. Methods: Ultra performance liquid chromatography(UPLC) was used to establish the fingerprints of Plantaginis root, Plantaginis leaf,Plantaginis semen and the remaining parts of the roots, leaves and seeds, the mobile phase was 0.2% methanoic acid in water(A) and acetonitrile(B) with gradient elution; DPPH·, ABTS+ were used to evaluate the anti-oxidation activity;LPS was used to induce Raw 264.7 cell inflammation model, and cell viability was measured by MTT assay. Levels of nitric oxide(NO) secreted by cells were determined by Griess to explore the anti-inflammatory activity and MTT were used to evaluate the anti-tumor activity. Results: The fingerprints of different medicinal parts of Plantaginis herba have similarities and some differences. Pharmacodynamic data showed that different medicinal parts of Plantaginis herba had good anti-inflammatory, anti-tumor and anti-oxidant activities, among which anti-inflammatory, anti-tumor activity was the highest in Plantaginis leaf, the remaining part of plantain is greater than psyllium; Anti-oxidant activity was the highest in the anterior leaf and low in the anterior root activity. Conclusion: This study clarified the fingerprints of alcohol extracts from different medicinal parts of Plantaginis herba and the differences in anti-inflammatory, anti-tumor and anti-oxidant activities, provided some data basis for further research on the pharmacodynamic substance basis of the different medicinal parts of Plantain and provide a basis for the efficient and rational use of medicinal resources of the Plantain and the development of new medicinal resources.
