玛咖对慢性束缚应激肝郁大鼠行为学及HPA轴的影响

摘要
目的:观察玛咖对慢性束缚应激(CRS)肝郁大鼠行为及下丘脑-垂体-肾上腺轴(HPA)的影响,探讨新"外来中药"玛咖疏肝解郁功效及作用机制。方法:将雄性SD大鼠根据蔗糖水消耗量及体质量随机分组,每组10只。除空白组外,其余均采用孤养结合CRS复制肝郁模型。观察体质量,行为学指标,并用放射免疫法(RIA)检测大鼠下丘脑促肾上腺皮质激素释放素(CRH),血浆促肾上腺皮质激素(ACTH)、皮质酮(CORT)的含量。结果:造模第21天,与模型组比较,玛咖醇提高剂量组体质量、糖水消耗量明显增加(P<0.05),旷场实验穿行次数明显增加(P<0.01,P<0.05),高架十字迷宫实验开放臂进入次数和停留时间明显增加(P<0.05),玛咖醇提高剂量组下丘脑CRH,血浆ACTH、CORT的含量明显降低(P<0.05,P<0.01)。结论:玛咖对慢性束缚应激致大鼠肝郁模型具有调节作用,其疏肝解郁功效可能与抑制肝郁模型大鼠HPAA的激活有关。
        Objective: To observe the effects of maca on behaviors and hypothalamic-pituitary-adrenal axis(HPAA) in chronic restraint stress(CRS) depressed rats and to explore the underlying mechanism. Methods: Male SD rats were randomly divided into eight groups according to the sucrose preference test and body weight(n=10). Except the normal control, the other groups were treated with the chronic stress stimulation combined with lonely. The behavior changes of rats were detected through open-field test, elevated plus-maze test, sucrose preference test and body weight were also measured. Radioimmunoassay(RIA) was used to analyze the levels of corticotrophin releasing hormone(CRH) in hypothalamus, adrenocorticotropic hormone(ACTH) and cortisol(CORT) in plasma. Results: Compared with model group, on the 21 th day, the weight and sugar consumption of high dose alcohol extract of maca group increased significantly(P<0.05). The cross times of open-field test of the maca group increased significantly(P<0.01, P<0.05). The times and stay period of entering open arms increased significantly(P<0.05), and the CRH, ACTH and CORT of the high dose alcohol extract of maca group decreased(P<0.05, P<0.01). Conclusion: Maca has a moderating effect on chronic restraint stress in stagnation of liver qi syndrome rats. Its function of soothing liver and relieving depression may be related to inhibiting the activation of HPAA in rats with liver stagnation.
