桑黄乙醇提取物的美白功效评价

摘要
探讨桑黄(Phellinus igniarius)乙醇提取物的美白功效。分别通过DPPH自由基清除实验、蘑菇酪氨酸酶抑制实验、B16F10黑色素瘤细胞胞内酪氨酸酶活性实验和B16F10黑色素瘤细胞黑色素含量测定实验评价桑黄乙醇提取物的美白活性。结果表明:桑黄乙醇提取物从1 mg/mL的浓度开始,对B16F10黑色素瘤细胞具有了极显著的细胞毒性,抑制细胞生长;其浓度为0.25、0.5 mg/mL时,对DPPH自由基的清除率分别达到了71.22%和80.94%,具有与维生素C接近的抗氧化活性;另外,其在浓度为0.25、0.5 mg/mL时,对蘑菇酪氨酸酶的抑制率分别达到了48.33%和65.12%,并且可显著降低B16F10黑色素瘤细胞胞内酪氨酸酶活性(P<0.01)和黑色素含量(P<0.01)。研究结果说明,桑黄乙醇提取物具有较强的美白功效。
        The whitening activity of a Phellinus igniarius ethanolic extract was studied. The whitening effect was assessed by the DPPH free radical scavenging assay, the mushroom tyrosinase actvitity assay and the tests of cell viability, intracellular tyrosinase activity and melanin content in B16 F10 cells. The results showed that Phellinus igniarius ethanolic extract had severe cytotoxicity and inhibited the growth of B16 F10 cells at a concentration of 1 mg/mL. At the concentrations of 0. 25 and 0. 5 mg/mL, the DPPH clearance rates reached 71.22% and 80.94% respectively, which were comparable to that of vitamin C. Moreover, at the concentrations of 0.25 and 0.5 mg/mL, the mushroom tyrosinase inhibition rates were 65.12% and 48.33%,and the intracellular tyrosinase activity and melanin content in B16 F10 cells were significantly reduced(P<0.01). These findings suggested that the ethanolic extract of Phellinus igniarius had a strong whitening effect.
