摘要
建立了一种测定制麦中阿魏酸含量的高效液相色谱法。选用色谱柱为Agilent Eclipse XDB-C18(4.6mm×250mm,5μm),流动相为甲醇-冰醋酸体系(60∶40,pH3.0),流速1.0ml/min,柱温35℃,紫外检测器波长320nm,进样量为20μl,对制麦中阿魏酸含量进行测定分析。结果表明:该方法条件下阿魏酸浓度在0.25~200μg/g时,线性关系良好(R2=0.999 9)。制麦过程中阿魏酸含量显著升高(P<0.05),麦芽阶段含量最高。所建方法分析速度快、操作简便、结果的精密度与重现性好,可作为制麦中阿魏酸含量的测定方法。
The study was to establish a HPLC method to determine the content of ferulic acid in malt.The chromatographic conditions were as follows:the chromatographic column was Agilent Eclipse XDB-C18(4.6 mm×250 mm,5μm),methanol-glacial acetic acid(60∶40,pH 3.0)as mobile phase with a flow rate of 1.0 ml/min,column temperature was 35℃,the detection wavelength was 320 nm andthe sample amount was 20μl.The method had good linear relationship when the quality ratio of ferulic acid was 0.25~200μg/g(R2=0.999 9).The content of ferulic acid during malting was significantly increased(P<0.05),which was the highest in malt.The method is more simple and faster,which could be used to determine the content of ferulic acid during malting for wheat.
