摘要
目的采用高效液相色谱法(HPLC)法建立铁皮石斛的指纹图谱,考察不同品种、产地和栽培基质铁皮石斛药材质量的差异情况。方法 HPLC法采用Zorbax SB-C18色谱柱(46 mm×250 mm,5μm),以0.1%磷酸溶液-乙腈为流动相,进行梯度洗脱,筛选检测波长为315 nm,测定了13批铁皮石斛样品的指纹图谱,并通过指纹图谱评价软件和聚类分析等手段,计算相似度,对样品进行评价。结果建立了铁皮石斛的指纹图谱,13批各类铁皮石斛样品共有15个共有峰,相似度评价结果显示:1批样品为0.565,其余均在0.704~0.966之间,聚类分析结果显示13批样品可以聚为三大类。结论建立的指纹图谱方法简便可靠,铁皮石斛的内在质量受品种、栽培基质的影响较大,受产地的影响相对较小。
        Objective To establish HPLC fingerprint for Dendrobium officinale and investigate the quality differences of Dendrobium officinale samples from different varieties,geographical origins and cultural substrates.Methods The HPLC separation was performed on a Zorbax(46 mm×250 mm,5μm)column with the mobile phase consisted of 0.1%phosphoric acid-acetonitrile,gradient elution,and the detection wavelength was 315 nm.The fingerprint of 13 batches of samples were tested.The similarity was calculated and the samples were evaluated by means of fingerprint evaluation software and cluster analysis.Results A fingerprint of Dendrobium officinale was established with 15 common peaks.Similarity evaluation results show that:except 1 batch of samples was 0.565,the others were between 0.704 and 0.966.Cluster analysis showed that 13 batches of samples could be grouped into three categories.Conclusion The established method of fingerprint is simple and reliable.The inner quality of Dendrobium officinale was greatly influenced by its variety and culture substrate,and was relatively less affected by the geographical origins.
