摘要
以1株野生侧耳属菌株为试验材料,采用ITS测序、同源序列比对、构建系统发育树进行鉴别,通过单因素试验确定影响其菌丝生长的因素。结果表明:该野生侧耳属菌株为哥伦比亚侧耳(Pleurotus columbinus),其菌丝生长的最佳碳源为蔗糖,最佳氮源为牛肉膏,最适碳氮比为10∶1,最适pH值为6.0,最适培养温度为25~30℃。
A wild Pleurotus strain was used as a test material. The wild strain was identified by ITS sequencing,homologous sequence alignment and phylogenetic tree construction. The biological characteristics were determined by single factor test.The results showed that the wild Pleurotus strain was identified as Pleurotus columbinus,the best carbon source was sucrose,the best nitrogen source was beef extract,the optimum carbon to nitrogen ratio was 10:1,the optimum pH value was 6.0,and the optimum culture temperature range was 25-30℃.
