摘要
目的:探讨红花蜂花粉多糖Ⅰ(PBPCⅠ)对小鼠肝癌H22实体瘤的抑制作用。方法:对肝癌H22实体瘤模型给予不同剂量的PBPCⅠ,采用ELISA法检测血清中IL-1α、IL-6、IFN-γ、TNF-α、IL-2、VEGF的含量,剥离瘤块,取脾脏及胸腺,称质量,计算抑瘤率与脏器指数。H&E染色法检测肿瘤细胞生长情况、肿瘤细胞形态及肿瘤细胞凋亡等指标。结果:与模型组相比,CTX组和PBPCⅠ低、中、高剂量组对小鼠H22肝癌的抑瘤率分别为67.93%、 33.48%、45.69%、57.86%。PBPCⅠ低、中、高剂量组能显著提高荷瘤小鼠体质量、免疫器官指数,并明显升高荷瘤小鼠血清中IFN-γ、IL-2、TNF-α含量,降低血清IL-1α、IL-6、VEGF含量。H&E染色发现不同浓度的PBPCⅠ均能使肝癌肿瘤细胞凋亡。结论:PBPCⅠ对H22荷瘤小鼠具有抑瘤作用,其作用可能与调节机体免疫功能有关。
        Objective:To investigate the inhibitory effect of safflower bee pollen polysaccharide I(PBPCI) on H22 solid tumor in mice.Methods:Liver cancer H22 solid tumor model was given different doses of PBPCI. The serum levels of IL-1α,IL-6, IFN-γ, TNF-α, IL-2, VEGF were measured by ELISA. The tumor were removed,the spleen and thymus were taken and weighed. The tumor inhibition rate and organ index were calculated. H&E staining method is used as detecting tumor cell growth,morphology and apoptosis indexes.Results:Compared with model group, CTX group and PBPCⅠlow, medium and high dose on H22 liver cancer in mice, the inhibitory rate of 67.93%, 33.48%, 45.69%, 57.86%. At the same time, PBPC Ⅰ low, medium and high dose group could significantly improve the body weight and immune organ index of tumor-bearing mice, and significantly increase the serum content of IFN-γ, IL-2 and TNF-α in tumor-bearing mice, reduce the content of IL-1α, IL-6, VEGF in serum.H&E staining showed that different concentrations of PBPCI caused apoptosis of liver cancer tumor cells.Conclusion:PBPCⅠ has a significant inhibitory effect on H22 mice, the effect may be associated with the immune function.
