甘露消毒丹及其挥发油对流感病毒感染小鼠黏膜免疫的影响

摘要
目的:观察甘露消毒丹(GXM)及其挥发油对流感病毒(IV)感染的BALB/c小鼠免疫球蛋白、细胞因子的含量变化以探讨其对黏膜免疫的影响。方法:SPF级BALB/c小鼠108只按照随机数字表法分为6组,分别为:正常组(A组)、模型组(B组)、GXM灌胃组(C组)、GXM滴鼻组(D组)、GXM合用组(E组)、奥司他韦组(F组),每组18只。除A组动物外,其余5组均以流感病毒FM1-6-E2滴鼻制备感染模型。于造模后第3、7、10天采集标本,采用ELISA方法检测肺泡灌洗液(BALF)及鼻咽冲洗液(NAIF)中sIgA、血清中IgA、IL-5、IL-6含量。结果:IV感染小鼠后,NAIF和BALF中sIgA含量降低;血清中IL-5、IL-6水平升高,而IgA含量降低。与B组比较,D、E组能够升高NAIF中sIgA含量,而C组在第10天升高明显(P<0.05),F组作用不明显(P>0.05);E组能够升高血清中IgA、BALF中sIgA含量,而C组、D组分别在第7天、第10天升高明显(P<0.05),F组作用不明显(P>0.05);C、E、F组能够降低IL-5、IL-6含量(P<0.05),而D组作用不明显(P>0.05)。结论:GXM挥发油可促进sIgA的分泌且起效迅速,GXM醇提物可升高IgA、sIgA含量,降低IL-5、IL-6含量,两种给药方式合用效果更佳。
        Objective:To observe the changes of immunoglobulin and cytokines in BALB/c mice infected with influenza viruses by using Ganlu Xiaodu Micropill(GXM) and its volatile oil to investigate its effect on mucosal immunity.Methods:Totally 108 SPF BALB/c mice were randomly divided into 6 groups according to random number table,respectively:normal group(Group A),model grape(Group B),filling GXM to the stomach group(Group C),dripping GXM to the nasal cavity(Group D),GXM combined group(Group E)and oseltamivir group(Group F),each group 18.In addition to the group A,we used nasalinoculation of IV FM1-6-E2 in mice to produce model.Each group's mice were infected on 3,7,10 day. And then mice were sacrificed,detecting the levels of sIgA in the miceBALF and NAIF,IgA,IL-5 and IL-6 in the mice bloodwith the method of ELISA.Results:After the infection of IV,SIgA levels decreased in NAIF and BALF;the levels of IL-5 and IL-6 in the mice blood were increased, but the level of IgA was decreased.Compared with group B,D, E group can increase the level of sIgA in NAIF,while that of the group C increased significantly on the 10 th day(P<0.05). The level of sIgA of F group was not obvious(P>0.05).E group can increase the levels of IgA, sIgA in BALF of C group and D group, respectively. On the 7 the and 10 the day,those increased significantly(P<0.05). Those of F group was not obvious(P>0.05). C, E, F group can reduce the levels of IL-5 and IL-6(P<0.05). D group'swere not obvious(P>0.05).Conclusion:GXM volatile oil can promote the secretion of sIgA and rapid onset. GXM alcohol extract can increasethe level of IgA and sIgA,reduce the levels of IL-5 and IL-6. The combination of the two model is better.
