摘要
目的:观察益智仁-乌药醚提取物对糖尿病肾病小鼠p27kip1蛋白表达的影响。方法:应用STZ造模,建立糖尿病肾病模型小鼠,分别给予厄贝沙坦10mg/(kg·d)、益智仁-乌药醚提取物3个剂量,即20g/(kg·d)、10g/(kg·d)、5g/(kg·d)治疗4周,采用RT-PCR和Western blotting方法观察各组小鼠p27kip1蛋白的表达情况。结果:与正常对照组比较,糖尿病肾病模型组p27kip1蛋白表达明显上调;与模型组比较,益智仁-乌药醚提取物20g/kg组和厄贝沙坦组p27kip1蛋白表达均明显下调。结论:益智仁-乌药醚提取物对糖尿病肾病小鼠肾脏有一定的保护作用,其机制可能与调节p27kip1蛋白表达有关。
Objective: To observe the influence of ether extract of the Yizhiren-Wuyao on p27kip1 Protein Expression in diabetic nephropathy mice.Methods: STZ was applied to establish the model of diabetic nephropathy mice. Diabetic nephropathy mice were given positive control drug irbesartan 10mg/( kg·d),ether extract of the Yizhiren-Wuyao three intervendonal 20 g/( kg·d),10 g/( kg·d),5g/( kg·d) for 4 weeks.The expression of p27kip1 Protein was observed by RT-PCR and Western blotting method. Results: Compared with the normal control group,diabetic nephropathy increased expression of p27kip1 Protein in the model group( P < 0. 01); compared with the model group,in the ether extract of the Yizhiren-Wuyao 20g/kg group and irbesartan group p27kip1 Protein expressions significantly decreased( P < 0. 01). Conclusion:ether extract of the Yizhiren-Wuyao on kidney of diabetic nephropathy mice has a protective effect,its mechanism may be related to regulating the expression of p27kip1 protein.
