通腑化浊凉血方对慢性肾衰竭模型大鼠硫酸吲哚酚清除的影响

摘要
目的:观察通腑化浊凉血方对于慢性肾衰竭模型大鼠硫酸吲哚酚清除的影响。方法:雄性SD大鼠60只随机分为空白组、模型组、通腑化浊凉血方低、中、高剂量组、尿毒清组。运用5/6肾切除法慢性肾衰竭大鼠模型,在造模成功后3个月对大鼠血肌酐、尿素氮、24 h尿蛋白含量、血液中硫酸吲哚酚含量进行检测,观察大鼠肾脏生化指标以及血清中肠源性尿毒症毒素硫酸吲哚酚含量情况。结果:与模型组相比,通腑化浊凉血方中、高剂量均可以降低模型大鼠血清BUN水平,明显降低SCr水平,三个剂量通腑化浊凉血方组大鼠24 h尿蛋白均有不同程度下降,血清IS水平较空白组明显上升。结论:通腑化浊凉血方可降低大鼠血清中硫酸吲哚酚含量,表明此方对此类蛋白结合型尿毒症毒素的清除具有一定优势,从而达到延缓肾损害进展的目的。
        Objective: To observe the effect of Tongfu Huazhuo Liangxue Formula on the elimination of indoxyl-sulfate(IS) in rats with chronic renal failure(CRF). Methods: 60 SD male rats were randomly divided into the blank group, the model group, the groups of Tongfu Huazhuo Liangxue Formula(low-dose, medium-dose, high-dose), and the Niaoduqing group. The CRF model was established by 5/6 nephrectomy. The levels of serum creatinine, urea nitrogen, 24-hour urinary protein and IS were measured, as well as the biochemical indexes of kidney and the content of intestinal uremic toxin IS were observed three months after successful modeling in rats. Results: The serum levels of BUN and SCr were significantly decreased in the high-dose group and the medium-dose group compared to those in the model group. The urinary protein was reduced to different degrees and serum level of IS was increased significantly in the groups of Tongfu Huazhuo Liangxue Formula with different doses. Conclusion: Tongfu Huazhuo Liangxue Formula can reduce the content of IS, which indicates that the formula has certain advantages in clearing protein-binding uremic toxins, thus to delay the progression of renal damage.
