摘要
目的探讨筋舒胶囊对小鼠的镇痛作用及对急性血瘀模型大鼠血液流变学的影响。方法将50只KM小鼠分为模型组(等体积纯化水),阳性药组(阿司匹林肠溶片61 mg/kg)及筋舒胶囊高、中、低剂量组(生药20. 8,10. 4,5. 2 g/kg),各10只,分别灌胃给予相应药物,每日1次,连续5 d;末次给药后1 h,各组小鼠均腹腔注射0. 8%冰醋酸0. 2 mL,记录注射后20 min内小鼠出现扭体反应的次数及潜伏期。70只SD大鼠分为空白对照组(等体积纯化水),模型组(等体积纯化水),阳性药组(复方丹参片1. 1 g/kg)及筋舒胶囊高、中、低剂量组(生药10. 4,5. 2,2. 6 g/kg),各10只,分别灌胃给予相应药物,每日1次,连续7 d;末次给药1 h后,除空白对照组外,其余各组大鼠均按0. 08 mL/100 g剂量皮下注射0. 1%肾上腺素,2 h后将大鼠置4℃冰水中浸泡5 min,于首次注射后4 h再次给予同剂量的肾上腺素,禁食不禁饮18 h;次日给予戊巴比妥钠45 mg/kg麻醉,采血,测定血液流变学指标。结果高、中剂量筋舒胶囊能显著延长冰醋酸所致小鼠扭体反应潜伏期和减少小鼠扭体反应次数(P <0. 01),低剂量仅能明显延长冰醋酸所致小鼠扭体反应潜伏期(P <0. 05);高、中剂量筋舒胶囊能显著降低急性血瘀模型大鼠的低切全血黏度和低切还原黏度(P <0. 05或P <0. 01)。结论筋舒胶囊对模型小鼠有明显的镇痛作用,对模型大鼠也有一定活血化瘀作用。
        Objective To investigate the analgesic effect of Jinshu Capsules on mice and their effect on hemorheology of acute blood-stasis model rats. Methods Totally 50 KM mice were divided into model group( equal volume of purified water),positive drug group( 61 mg/kg Aspirin Enteric-Coated Tablets) and Jinshu Capsule high,medium and low dosage groups( equivalent to 20. 8,10. 4,5. 2 g/kg crude drug),10 mice in each group,they were given intragastric administration of corresponding drugs once a day for 5 consecutive days. 1 h after the last administration,the mice in each group were intraperitoneally injected with 0. 8% glacial acetic acid( 0. 2 m L),the times of writhing reactions and their latency were recorded within 20 min after the injection. Totally 70 SD rats were divided into blank control group( equal volume of purified water),model group( equal volume of purified water),positive drug group( 1. 1 g/kg Compound Danshen Tablets) and Jinshu Capsules high,medium and low dosage groups( equivalent to 10. 4,5. 2,2. 6 g/kg crude drug),10 rats in eacch group,they were given intragastric administration of corresponding drugs once a day for 7 consecutive days. 1 h after the last administration,except for the blank control group,the rats in the other groups were subcutaneously injected with 0. 1%epinephrine at a dosage of 0. 08 mL/100 g. After 2 h,the rats were immersed in 4 ℃ ice water for 5 min,the rats were given the same dosage of epinephrine again at 4 h after the first injection,and then fasting without liquid fasting for 18 h. On the next day,rats were anesthetized with 45 mg/kg pentobarbital sodium,and the blood samples were collected to determine the hemorheological parameters.Results High and medium dosage of Jinshu Capsules could significantly prolong the latency of writhing reaction induced by glacial acetic acid in mice and reduce the times of writhing reaction in mice( P < 0. 01). Low dosage of Jinshu Capsules could significantly prolong the latency of writhing reaction induced by glacial acetic acid in mice( P < 0. 05). High and medium dosage of Jinshu Capsules could significantly reduce the low shear whole blood viscosity and low shear reduction viscosity in acute blood-stasis model rats( P < 0. 05 or P < 0. 01). Conclusion Jinshu Capsules have obvious analgesic effect on model mice and has certain effect of activating blood circulation and removing blood stasis on model rats.
