柴芍胃炎颗粒对胃溃疡大鼠黏膜细胞因子的影响

摘要
目的:研究柴芍胃炎颗粒对胃溃疡愈合的影响,并探讨其机制。方法:40只wistar大鼠,随机分为正常组、模型组、奥美拉唑组、柴芍胃炎颗粒组(简称胃炎颗粒组),采用改良Takagi法制作乙酸胃溃疡大鼠模型,治疗13天后测量各组溃疡指数,用免疫组化法检测胃黏膜细胞Bcl-2、NF-κB蛋白的阳性密度。结果:柴芍胃炎颗粒组与其他组比较溃疡指数明显降低(P<0.01),柴芍胃炎颗粒组与奥美拉唑组Bcl-2的表达明显高于模型组(P<0.01),且柴芍胃炎颗粒组高于奥美拉唑组(P<0.01)。柴芍胃炎颗粒组NF-κB的表达明显低于模型组(P<0.01)。结论:柴芍胃炎颗粒能通过提高Bcl-2抑制细胞凋亡、降低NF-κB蛋白的表达促进溃疡愈合,并可能是其促进溃疡愈合机制之一。
        Objective:To study the protective effect of Chaishao Weiyan Granules on gastric ulcer of rats and its effective mechanism. Method:40 healthy wistar rats were randomly divided into model group,Chaishao Weiyan Granules group,omeprazole treatment group and normal group. The rat model of gastric ulcer was made by Takagi's method with some modification. After the rats were treated for 13 d,the index of each group of ulcer and the expression of Bcl-2 and NF-κB in the gastric ulcer tissues was examined by immunohistochemical method. Result:Chaishao Weiyan Granules group could obviously decrease the ulcer index more than the others group(P<0.01). Compared with that of the model group,the expression of Bcl-2 in Chaishao Weiyan Granules group and omeprazole group was significantly higher(P<0.01)and that of Chaishao Weiyan Granules group was higher than that of omeprazole group(P<0.01).The expression of NF-κB Chaishao Weiyan Granules group was significantly lower than that of the model group(P<0.01).Conclusion:Chaishao Weiyan Granules could increase Bcl-2 to inhibit apoptosis and reduce the expression of NF-κB protein to promote ulcer healing,which may be one of the mechanisms to promote ulcer healing.
