丁苯酞对脑缺血再灌注大鼠脑红蛋白和14-3-3γ蛋白的影响

摘要
目的探讨丁苯酞对脑缺血再灌注大鼠脑组织和血清脑红蛋白(Ngb)和14-3-3γ蛋白水平的影响。方法将30只雄性SD大鼠随机分为假手术组、缺血再灌注组和丁苯酞组,建立大鼠大脑中动脉缺血再灌注模型;24 h后采集大脑和血清,苏木素-伊红(HE)染色观察梗死灶周围的细胞损伤,采用免疫组化、酶联免疫吸附试验(ELISA)分别检测大脑和血清Ngb和14-3-3γ水平。结果 HE染色显示丁苯酞组梗死灶周围细胞损害较缺血再灌注组显著减轻,缺血再灌注组脑组织和血清14-3-3γ和Ngb表达均显著高于假手术组(P<0.05),而丁苯酞组脑组织和血清14-3-3γ和Ngb水平均显著高于缺血再灌注组(P<0.05)。结论丁苯酞减轻脑缺血再灌注大鼠脑梗死灶周围再灌注损害,其作用机制之一是上调脑组织和血清14-3-3γ和Ngb水平。
        Objective To explore the effect of butylphthalide on the neuroglobin(Ngb) and 14-3-3γ levels of cerebra and serum in rats with middle cerebral artery ischemia-reperfusion.Methods 30 male SD rats were randomly divided into sham operation, ischemia-reperfusion and butylphthalide-pretreated groups. The cerebral ischemia-reperfusion rat model was established in ischemia-reperfusion and butylphthalide-pretreated group. 24 hours later, the brain and serum were collected and the expressions of Ngb and 14-3-3γ were detected by ELISA and immunohistochemistry.Results The Ngb and 14-3-3γ levels of cerebra and serum were higher in ischemia-reperfusion group compared to those of sham operation group(P<0.05), while Ngb and 14-3-3γ expressions were higher in butylphthalide-pretreated group than those in ischemia-reperfusion group(P<0.05).Conclusions Butylphthalide could protect against middle cerebral artery ischemia-reperfusion in rats through upregulating production of Ngb and 14-3-3γ.
