程氏蠲痹汤对类风湿关节炎SD大鼠血清白细胞介素-17和-35水平的影响

摘要
目的探求程氏蠲痹汤治疗类风湿关节炎的作用机制及发挥良效的剂量和时间。方法以程氏蠲痹汤高、低剂量治疗类风湿关节炎SD大鼠,并与雷公藤多苷片组、模型组、正常组比较,观察治疗第7、14、21天各组血清中白细胞介素(IL)-17和IL-35的含量差异,IL-17、IL-35两者变化的相关性及在3个时间点各组关节、肝肾等的病理改变。结果治疗第14天,程氏蠲痹汤高剂量组IL-17、IL-35水平与正常组无明显差异(P>0.05),雷公藤多苷片组IL-17、IL-35水平与正常组差异明显(P<0.05);治疗第21天,程氏蠲痹汤高剂量组、雷公藤多苷片组与正常组IL-17、IL-35水平无明显差异(P>0.05);在3个时间点,程氏蠲痹汤低剂量组与模型组无明显差异(P>0.05),模型组与正常组差异显著(P<0.01),各组关节病理改变与血清IL-17、IL-35含量变化一致,程氏蠲痹汤高剂量组、雷公藤多苷片组肝肾病理改变与正常组一致;程氏蠲痹汤高剂量组IL-17、IL-35血清含量第7～14天呈负相关(P<0.05)。结论程氏蠲痹汤高剂量有效提升血清IL-35含量,抑制IL-17释放,从而发挥抑炎疗效,比常用药雷公藤多苷片更早达良效并维持。
Objective To investigate the influence of Cheng Juanbi Decoction on serum levels of IL-17 and IL-35 in SD rats with rheumatoid arthritis.Methods Rheumatoid arthritis SD rats were treated with high and low doses of Cheng Juanbi Decoction. The levels of interleukin(IL)-17, IL-35 were tested in high and low dose Cheng Juanbi Decoction, tripterygium glycoside treatment, model and normal groups.The pathological changes of joints, liver and kidney in each group were observed at different time points.Results At the 14 th of treatment, compared with those of normal group, the levels of IL-17, IL-35 in Cheng Juanbi Decoction high dose group had no obvious differences(P>0.05), the levels of IL-17,IL-35 in tripterygium glycoside treatment group had obvious differences compared with those of normal group(P<0.05).At the 21 st of treatment, compared with those of normal group, the levels of IL-17, IL-35 in Cheng Juanbi Decoction high dose group and tripterygium glycoside treatment group had no obvious differences(P>0.05). At three time points, there were no obvious differences between Cheng Juanbi Decoction low dose group and model group(P>0.05), and there were obvious differences between model group and normal group(P<0.01).Pathological changes of joints in each group were in line with changes of serum IL-17 and IL-35 contents in each group. Pathological changes of liver and kidney in Cheng Juanbi Decoction high dose group and tripterygium glycoside group were in line with those in normal group.There was the negative correlation between the content of IL-17 and IL-35 in Cheng Juanbi Decoction high dose group from 7 th to 14 th day.Conclusions The high dose Cheng Juanbi Decoction could enhance serum content of IL-35 effectively and restrain release of IL-17 and thus exert anti inflammatory effects.Compared with tripterygium glycoside, the high dose Cheng Juanbi Decoction could achieve good effect, low dose Cheng Juanbi Decoction has no therapeutic significance.
