摘要

目的研究阿霉素脂质体(DOX-Lip)对人前列腺癌细胞裸鼠移植瘤生长的抑制作用。方法将Caco-2细胞分为空白组(0. 9%Na Cl)、对照组(10μg·m L~(-1)DOX)和实验组(10μg·m L~(-1)DOX-Lip),用流式细胞术检测细胞摄取。将裸鼠人前列腺癌移植瘤模型裸鼠随机分为空白组、对照组和实验组。对照组和实验组分别为予以10μg·m L~(-1)DOX和10μg·m L~(-1)DOX-Lip,空白组给予同等体积的0. 9%Na Cl。3组裸鼠均隔天灌胃给药1次,干预24 d。实验结束后称量瘤重,用TUNEL分析肿瘤细胞凋亡情况。结果与空白组Caco-2细胞的平均荧光强度值(6. 8±0. 9)比较,实验组和对照组Caco-2细胞的平均荧光强度值分别为(664. 1±9. 5)和(466. 2±12. 9),差异均有统计学意义(均P <0. 01),且对照组和实验组的平均荧光强度值比较,差异有统计学意义(P <0. 05)。实验组、对照组和空白组的瘤重分别为(0. 08±0. 02),(0. 39±0. 03)和(0. 68±0. 05) g,凋亡率分别为(32. 34±4. 27)%,(12. 83±1. 25)%和(0. 98±0. 12)%,实验组的上述指标与对照组比较,差异均有统计学意义(均P <0. 05)。结论 DOX-Lip对人前列腺癌细胞裸鼠移植瘤生长有较好的抑制作用。
        Objective To study the inhibitory effect of doxorubicin liposome( DOX-Lip) on the growth of human prostate cancer xenografts in nude mice. Methods The cells were divided into blank group( 0. 9%NaCl),control group( 10 μg·m L~(-1) DOX) and experimental group( 10μg·m L~(-1) DOX-Lip). The detection of cell uptake was performed by flow cytometry. The nude mice models of human prostate cancer transplantation were randomly divided into blank,control and experimental groups. The control, experimental and blank groups were given 10μg·m L~(-1) DOX,10 μg · m L~(-1) DOX-Lip and the equivalence of0. 9% NaCl,respectively. Three groups were gavage administration for 24 days with once every other day. After the experiment,the tumor weight was weighed and the apoptosis of tumor cells was analyzed by TUNEL method. Results Compared with blank group( 6. 8 ± 0. 9),the mean fluorescence intensity values of experimental and control groups were( 664. 1 ± 9. 5) and( 466. 2 ± 12. 9) with significant differences( all P < 0. 05). Also,there was statistically significant difference between experimental group and control group( P < 0. 05). The tumor weights of the experimental,control and blank groups were( 0. 08 ± 0. 02),( 0. 39 ± 0. 03) and( 0. 68 ± 0. 05) g,and the apoptotic rates were( 32. 34 ± 4. 27) %,( 12. 83 ± 1. 25) % and( 0. 98 ± 0. 12) %,the differences were statistically significant between experimental group and control group( P < 0. 05). Conclusion DOX-Lip has a good inhibitory effect on the growth of human prostate cancer cells in nude mice.
