摘要
目的:探讨丝胶对2型糖尿病大鼠胰腺磷脂酰肌醇-3-激酶(phosphatidylinositol3-kinase,PI3K mRNA表达的影响。方法:雄性SD大鼠随机分为3组,正常对照组、糖尿病模型组、丝胶治疗组。以高脂高糖饲料喂养联合链脲佐菌素(35mg/kg,2次)注射的方法建立2型糖尿病大鼠模型,丝胶治疗组大鼠给予丝胶(2.4g/kg/d)灌胃。采用葡萄糖氧化酶法检测每组大鼠的血糖,Real Time Q-PCR法检测每组大鼠胰腺PI3K mRNA的表达。结果:与正常对照组比较,糖尿病模型组大鼠的血糖明显升高,胰腺PI3K mRNA的表达明显降低(P<0.05);与糖尿病模型组比较,丝胶治疗组大鼠的血糖明显降低,胰腺PI3KmRNA的表达明显升高(P<0.05)。结论:丝胶可能通过提高胰腺PI3K的表达调节胰岛素PI3K-Akt信号通路,从而起到降低血糖的作用。
        Objective: To investigate the effects of sericin on phosphatidylinositol-3-kinase(PI3 K) mRNA expression in pancreas of type 2 diabetic rats. Methods: Male SD rats were randomly divided into three groups: normal control group,diabetic model group and sericin treatment group. The rat model of type 2 diabetes mellitus was established by feeding high-fat and high-sugar diet and intraperitoneal injection of streptozotocin(35 mg/kg, twice). Then the rats in the sericin treatment group were given sericin(2.4 g/kg/d) by gavage. Glucose oxidase method was used to detect the blood glucose of rats, and Real Time Q-PCR were used to detect the expression of PI3 K mRNA in pancreas of rats in each group. Results:Compared with the normal control group, the blood glucose of diabetic rats increased significantly and the expression of PI3 K mRNA in pancreas decreased significantly(P<0.05); Compared with diabetic rats, the blood glucose of sericin treated rats decreased significantly, and the expression of PI3 K mRNA in pancreas increased significantly(P<0.05). Conclusions:Sericin may regulate insulin PI3 K-Akt signaling pathway by increasing the expression of PI3 K in pancreas, thus reducing blood glucose.
