柴胡皂苷对癫痫大鼠皮质和海马区谷氨酸、γ-羟基丁酸含量的影响

摘要
目的观察柴胡皂苷对癫痫大鼠皮质和海马区谷氨酸、γ-羟基丁酸含量调节的影响。方法将健康SD大鼠给予反复腹腔注射氯化锂-匹罗卡品化学点燃建立大鼠癫痫的动物模型,将制备成功的癫痫大鼠模型随机分为模型对照组及柴胡皂苷组,各15只,另选择15只正常大鼠作为正常对照组。柴胡皂苷组予以腹腔注射柴胡皂苷悬浊液1.09 mL/(kg·d),模型对照组及正常对照组给予等量0.9%氯化钠,使用方法、剂量及使用频率和柴胡皂苷组相同至实验结束。比较各组大鼠癫痫发作情况、皮质和海马区谷氨酸和γ-羟基丁酸含量。结果治疗后4周、8周,与模型对照组比较,柴胡皂苷组癫痫发作次数减少、发作时间短,且柴胡皂苷组发作次数、发作时间持续降低(P <0.05)。与正常对照组比较,模型对照组和柴胡皂苷组皮质和海马区谷氨酸阳性细胞数较高(P <0.05);与模型对照组比较,柴胡皂苷组皮质和海马区谷氨酸阳性细胞数较少(P <0.05)。治疗后,与正常对照组比较,模型对照组及柴胡皂苷组皮质和海马区γ-羟基丁酸阳性细胞数较少(P <0.05);与模型对照组比较,柴胡皂苷组皮质和海马区γ-羟基丁酸阳性细胞数较多(P <0.05)。结论柴胡皂苷可降低癫痫大鼠皮质和海马脑区兴奋性氨基酸谷氨酸水平,减缓病变神经元兴奋性,起到抗癫痫作用,提高γ-羟基丁酸含量,降低神经元兴奋性,抑制癫痫发作,缓解癫痫发作所致的神经系统受损,具有显著抑制癫痫发作、保护神经元作用。
        Objective To investigate the effects of Radix Bupleuri(RB)on the contents of glutamate and gamma-aminobutyric acid(GABA)in cortex and hippocampus of epileptic rats.Methods Healthy Sprague-Dawley(SD)rats were given repeated intraperitoneal injection of lithium pilocarpine kindling to establish the rat models of epilepsy.The model rats were randomly divided into model control group(n=15)and RB group(n=15),and 15 normal rats were selected as normal control group.The rats in RB group were received intraperitoneal injection of RB1.09 mL/(kg·d).The rats in control group and the normal control group were given the same amount of 0.9% sodium chloride.The epileptic seizures,the contents of glutamate,and GABA in cortex and hippocampus of rats in each group were compared.Results After treatment for 4 weeks and 8 weeks,compared with the model control group,the numbers of seizures and the average time of onset in RB group were lower,and the numbers of seizures and the average duration of the onset were decreased(P <0.05).Compared with the normal control group,the number of glutamate positive cells in the cortex and hippocampus were higher in the model group and RB group(P <0.05).Compared with the model group,the number of glutamate positive cells in the cortex and hippocampus was lower in RB group(P <0.05).After treatment,compared with the normal control group,GABA positive cells in cortex and hippocampus was lower in model control group and RB group(P <0.05).Compared with the model group,GABA positive cell count was higher in cortex and hippocampus of RB group(P <0.05).Conclusion RB can reduce the level of excitatory amino acid glutamate in the cortex and hippocampus of rats with epilepsy,slow down the excitability of diseased neurons,play an anti-epileptic effect,increase the content ofγ-hydroxybutyric acid,reduce the excitability of neurons,and inhibit epilepsy attack,relieve the damage of the nervous system caused by seizures inhibit of seizures and protect neurons.
