摘要
目的:观察麦冬皂苷B(O P-B)对人胃癌M G C-803细胞侵袭和迁移能力的影响及其作用机制初探。方法:体外培养人胃癌MGC-803细胞,分别以0、0.1、1、5、10、15和20μmol/L的OP-B干预MGC-803细胞24、48h,利用CCK-8法观察OP-B对胃癌MGC-803细胞活性的影响;细胞侵袭实验和划痕实验检测OP-B(5、10、15μmol/L)作用MGC-803细胞24h后细胞的侵袭和迁移能力。蛋白免疫印迹法(WesternBlot)检测细胞中MMP-2、MMP-9蛋白水平的变化。结果:OP-B能明显抑制胃癌MGC-803细胞增殖(P<0.05),并抑制其侵袭和迁移(P<0.05),同时能降低其MMP-2、MMP-9蛋白表达水平(P<0.05)。结论:OP-B能够抑制人胃癌MGC-803细胞的侵袭和迁移能力,其作用机制可能与抑制MMP-2、MMP-9蛋白的表达有关。
Objective: To observe the effects of ophiopogonin B(OP-B) on the invasion and migration of human gastric cancer MGC-803 cells, and to explore the possible mechanism. Methods: MGC-803 cells were treated with different concentrations(0, 0.1, 1, 5, 10, 15 and 20μmol/L) of OP-B for 24 h and 48 h. CCK-8 was used to detect the inhibitory effect of OP-B on the proliferation inhibition rate of MGC-803 cells. Transwell and scratch test were used to detect the invasion and migration ability of MGC-803 cells after treated with OP-B(5, 10, 15μmol/L) for 24 h. Western Blot was used to detect the expression levels of MMP-2 and MMP-9. Results: OP-B significantly inhibited the proliferation of gastric cancer MGC-803 cells(P<0.05), and reduced the invasion and migration ability of MGC-803 cells(P<0.05). In addition, 15μmol/L OP-B could decrease the expression levels of MMP-2 and MMP-9 in MGC-803 cells(P<0.05). Conclusion: OP-B can inhibit the proliferation, invasion and migration ability of MGC-803 cells. The mechanism may be related to the inhibition of MMP-2 and MMP-9 protein expression.
