地蜂子中儿茶素含量测定及其醇浸物对软组织损伤的影响

摘要
目的:建立地蜂子中儿茶素含量测定方法,观察地蜂子醇浸物对软组织损伤小鼠的治疗效果。方法:采用薄层色谱法(TLC)对药材进行定性鉴别,用高效液相色谱法(HPLC)测定地蜂子中儿茶素的含量;采用砝码法制作30只小鼠软组织损伤模型,将模型小鼠均分为空白组(涂抹生理盐水)、阳性对照组(涂抹云南白药)及地蜂子组(涂抹地蜂子液),连续给药3 d,记录小鼠损伤症候指数、观察治疗效果。结果:TLC斑点清晰,重复性好;地蜂子中儿茶素线性范围为0.017~0.086 g/L,r=0.999 4,回收率(n=6)为100.13%(RSD=2.18);与空白组比较,给药后第3天地蜂子组小鼠的损伤症候指数显著降低(P<0.05或P<0.01),损伤局部消肿效果较好。结论:本研究建立的高效液相色谱(HPLC)测定方法简单、准确,可用于地蜂子中儿茶素含量的测定;地蜂子醇浸物对小鼠软组织损伤具有改善作用。
        Objective: To establish stable determination method of catechin in Potentilla freyniana, and to observe the therapeutic effect of Potentilla freyniana on acute soft tissue injury. Method: TLC was adopted for qualitative identification of samples, and HPLC method was used to determine the content of catechin.The mice model of acute soft tissue injury were established by weight blow method, mouse were randomized into three groups and given different drugs. The mice were treated for 3 consecutive days, and the injury symptom score was observed and using SPSS17.0 software to compare the differences between groups for analysis of variance. Results: TLC spots of samples were clear and well repeatability. The linear range of catechin of Potentilla freyniana was 0.017~0.086 mg/mL, the correlation coefficient was 0.999 4,and the recovery rate was 100.13%(n=6,RSD=2.18). Compared with the blank group, the score of soft tissue injury in mice decreased significantly on the third day after treatment. Conclusion: The HPLC method established in this experiment is simple and accurate, and can be used for the determination of catechin in Potentilla freyniana and extract of Potentilla freyniana can diminish the soft tissue injury in mice.
