摘要
【目的】利用外源海藻糖,提高菌种抗高温能力,筛选耐高温品种。【方法】在培养基中添加外源海藻糖生产双孢蘑菇(Agaricus bisporus)菌种,提高抗高温势能,并根据不同品种在高温下的生长表现筛选耐高温品种(株系)。【结果】在添加外源海藻糖的培养基中,绝大多数品种在高温下菌丝生长快、生长势强且丰厚。供试各品种的生长特性可分3个类型:第1种类型是接种后菌丝萌发较快,第1周迅速生长,第2~3周生长缓慢,如Baichagu、2796系列的2796-Gaoyou、2796-4-9、Accc系列的Accc-52327、Accc-50842-411等品种;第2种类型品种接种后菌丝萌发较慢,第1周生长较缓,第2周迅速生长,如U3-4-22、日本引进品种JP-100和JP-118、F56系列;第3种类型是第1周菌丝体基本上不萌发,而在第2周后期,特别是第3周开始加快生长速度,如Accc-50554、Accc-50442-515、Mohuanong-2796-1等。【结论】筛选株系的结果表明,F56的2个株系,在小麦和玉米粒2种培养基质中生长良好,特别是在玉米粒培养基质中满袋率高达100%。2796-Gaoyou的株系,在小麦培养基质中满袋率达50%~85%以上,Ac-cc-50554的Accc-50554-1株系满袋率高达100%,其他所有株系也都在70%以上。
        【Objective】The present paper used exogenous trehalose to improve the ability of the strain to resist high temperature and to select the high temperature resistant varieties.【Method】The exogenous trehalose was added to cultivate Agaricus bisporus strain,the high temperature resistance was enhanced,and high temperature resistant varieties( strain) according to the growth performance of different varieties at high temperature were screened.【Result】The results showed that in the medium adding exogenous trehalose,most of the varieties showed that the growth of mycelium was fast,strong and rich at high temperature. The growth characteristics of the tested varieties can be divided into three types: The first types were rapid growth of mycelium after inoculation,rapid growth in first weeks and slow growth in second and third weeks,such as Baichagu,2796 group of 2796-Gaoyou,2796-4-9,Accc group of Accc-52327,Accc-50842-411 and other varieties; The second types were slow growth of mycelium after inoculation,slow growth in first weeks and rapid growth in second weeks,for example,U3-4-22,F56 series,Japanese imported varieties JP-100 and JP-118; The third type is that the mycelium does not germinate in the first week,and the growth speed is accelerated in the late second weeks,especially in the third week,such as Accc-50554,Accc-50442-515,Mohuanong-2796-1 and so on. 【Conclusion】The results of screening strains showed that two lines of F56 in wheat and corn grain,two kinds of culture medium are growing well,especially in corn culture medium,pocketfuls rate as high as 100 %; 2796-Gaoyou strains in wheat medium,rate of more than 50 %-85 % level; In the Accc-50554 series,Accc-50554-1 strain pocketfuls rate as high as 100 %,all other lines are in more than 70 %.
