风湿宁胶囊对寒湿闭阻型类风湿关节炎模型大鼠作用机制研究

摘要
目的:观察风湿宁胶囊对类风湿关节炎(寒湿闭阻)大鼠的治疗作用及机制。方法:雄性SD大鼠72只,随机分为6组,每组12只,采用牛II型胶原结合人工气候箱对SD大鼠造模,造模完成后对其进行灌胃给药,每日2次,连续28天,观察药物对大鼠关节容积、关节病理以及外周血中IL-1β、IFN-γ、IL-10、IL-4、IL-17a、TGF-β1等细胞因子的表达情况。结果:风湿宁胶囊可有效降低类风湿关节炎(寒湿闭阻)大鼠关节容积,显著减少滑膜组织中浸润的炎细胞及新生血管翳,明显调节外周血中炎性及抗炎因子的比例。结论:风湿宁胶囊对类风湿关节炎(寒湿闭阻)大鼠关节炎症有明显的治疗作用,其机制可能与改善了炎性及抗炎因子失衡的比例有关。
        Objective: To observe the therapeutic effect and mechanism of Fengshining Capsule on CIA model rats with cold damp stagnation syndrome. Methods: 72 male SD rats were randomly divided into 6 groups,12 rats in each group. SD rats were modeled by bovine type II collagen combined with artificial climate box. After modeling,they were administered gavage,2 times / d. After administration for 28 days,the expressions of drugs on rat's articular volume,joint pathology and IL- 1β,INF- γ,IL- 10,IL- 4,IL17- a,TGF- β1 etc. cytokines in peripheral blood were observed. Results: Fengshining Capsule could effectively reduce the articular volume of CIA rats with cold damp stagnation syndrome,significantly decrease the infiltrating inflammatory cells in synovial tissue and the neovascularization nebula,and obviously regulate the proportion between inflammatory and anti- inflammatory cytokines in peripheral blood. Conclusion: Fengshining Capsule has the obvious therapeutic effect on joint inflammation in CIA rats with cold dampness closed stagnation syndrome. Its mechanism may be related to the improvement of imbalance proportion between inflammatory and anti- inflammatory cytokines.
