摘要
目的通过辛伐他汀对豚鼠哮喘模型血清中IL-4、TGF-β1水平的影响,为辛伐他汀医治哮喘提供实验依据。方法 50只健康豚鼠被随机分为5组:哮喘模型组、S1:20mg/kg辛伐他汀干预组、S2:40 mg/kg辛伐他汀干预组、S3:60 mg/kg辛伐他汀干预组、对照组;每组10只。采用卵蛋白致敏和反复激发制备豚鼠哮喘模型。血清中IL-4、TGF-β1的水平用酶联免疫吸附法(ELISA)检测。结果哮喘模型组、S1组、S2组及S3组IL-4、TGF-β1水平均高于对照组(P<0.01)。其中,哮喘模型组高于辛伐他汀干预组(P <0.05);S1、S3组高于S2组(P <0.05)。结论辛伐他汀可通过降低IL-4水平减轻气道炎症;辛伐他汀可通过降低TGF-β1水平以发挥抑制气道重塑的作用。
        Objective To study the impact of simvastatin on serum IL-4,TGF-β1 in the asthma models guinea pigs, to provide the experimental basis for simvastatin treatment of bronchial asthma. Methods Fifty male guinea pigs were randomly divided into five groups: asthmatic model group, S1 group(20 mg/kg simvastatin treat group), S2 group(40 mg/kg simvastatin treat group) and S3 group(60 mg/kg simvastatin treat group) and control group, 10 guinea pigs in each group. The asthmatic models were established by ovalbumin(OVA) sensitization and repeated stimulation. Serum level of IL-4 and TGF-β1 were detected by enzyme-linked immunosorbent assay(ELISA). Results Compared with the control group, the level of IL-4, TGF-β1 in the asthma model group,S1 group,S2 group and S3 group were significantly higher(P < 0.01). The level of IL-4, TGF-β1 in asthma group was higher than simvastatin treat group(P < 0.05). The level of IL-4, TGF-β1 in S1 and S3 group was higher than S2 group(P < 0.05). Conclusion Simvastatin can reduce airway inflammation by lowering the level of IL-4. Simvastatin can inhibit asthma airway remodeling by reducing the level of TGF-β1.
