摘要
目的研究理气散结颗粒对大鼠乳腺增生的影响。方法 96只大鼠随机分为空白对照组、模型对照组、他莫昔芬组(0. 003 3 g/kg)、小金丸组(0. 200 0 g/kg)、理气散结丸组(1. 957 0 g/kg)及理气散结颗粒低、中、高剂量组(0. 978 5、1. 957 0、3. 914 0 g/kg),每组12只,除空白对照组外各组大鼠肌内注射苯甲酸雌二醇和黄体酮以诱导乳腺增生。给药30 d后取血,ELISA法测定血清雌二醇(E2)、孕酮(P)水平,免疫组化法检测乳腺组织雌激素受体(ER)、孕激素受体(PR)蛋白表达,光镜下观察乳腺组织病理变化。结果与模型对照组比较,理气散结颗粒中剂量组ER蛋白表达显著降低(P<0. 05);高剂量组E2水平显著升高(P<0. 05),ER、PR蛋白表达显著降低(P<0. 05,P<0. 01);低剂量组组织增生明显改善(P<0. 05)。结论理气散结颗粒对大鼠乳腺增生的影响可能与改善乳腺组织中ER、PR蛋白表达有关。
        AIM To study the effects of Liqi Sanjie Granules on mammary gland hyperplasia in rats.METHODS Ninety-six rats were randomly divided into blank control group,model control group,tamoxifen group( 0. 003 3 g/kg),Xiaojin Pills group( 0. 200 0 g/kg),Liqi Sanjie Pills group( 1. 957 0 g/kg) and Liqi Sanjie Granules low-dose,medium-dose,high-dose groups( 0. 978 5,1. 957 0,3. 914 0 g/kg),12 rats in each group,the rats in various groups( except for the blank control group) were given intramuscular injection of benzoate estradiol and progesterone to induce mammary gland hyperplasia. Blood collection was made after 30-day administration,serum estradiol( E2) and progesterone( P) levels were determined by ELISA,estrogen receptor( ER)and progesterone receptor( PR) protein expressions in mammary gland tissue were detected by immunohistochemistry,and the pathological changes of mammary gland tissue were observed under light microscope. RESULTS Compared with the model control group,the Liqi Sanjie Granules medium-dose group demonstrated significantly decreased ER protein expression( P<0. 05),the Liqi Sanjie Granules high-dose group exhibited markedly increased E2level( P<0. 05) with obviously reduced ER and PR protein expressions( P<0. 05,P<0. 01),and obviously improved tissue hyperplasia( P<0. 05) was observable in the Liqi Sanjie Granules low-dose group. CONCLUSION The effects of Liqi Sanjie Granules on mammary gland hyperplasia in rats may contribute to improving ER and PR protein expressions.
