摘要
目的检测Caspase-3、Caspase-8在非酒精性脂肪性肝炎中的表达,探讨其在肝炎中的临床意义。方法应用免疫组化方法和原位末端标记技术(TUNEL实验)分别检测50份肝炎肝组织和15份正常肝组织中Caspase-3、Caspase-8表达和细胞凋亡。结果 Caspase-3、Caspase-8在非酒精性脂肪性肝炎中的表达(82.0%、82.0%)分别高于其在正常肝组织中的表达(18.8%、13.3%),差异有统计学意义(χ2=20.281,χ2=24.298,P<0.05);并与肝细胞凋亡密切相关(r=0.356,r=0.409,P<0.001)。Caspase-3、Caspase-8在非酒精性脂肪性肝炎随病理损伤程度加重阳性表达率增高,差异有统计学意义(χ2=6.978,χ2=11.043,P<0.05)。Caspase-3、Caspase-8在非酒精性脂肪性肝炎中的表达与谷丙转氨酶水平比较差异无统计学意义(P>0.05)。结论 Caspase-3、Caspase-8参与非酒精性脂肪性肝炎肝细胞凋亡的发生,并参与非酒精性脂肪性肝炎的发病过程。
        Objective To detect the expression of Caspase-3 and Caspase-8 in non-alcoholic steatohepatitis and explore its clinical significance in hepatitis. Methods Immunohistochemistry and in situ end labeling(TUNEL test) were used to detect the expression of Caspase-3,Caspase-8 and apoptosis in 50 cases of hepatitis and 15 cases of normal liver tissues respectively. Results The expression of Caspase-3,Caspase-8 in non-alcoholic steatohepatitis(82. 0%,82. 0%) was higher than that in normal liver tissues(18. 8%; 13. 3%),and the difference was statistically significant(χ2= 20. 281,χ2= 24. 298,P <0. 05),and it is closely related to hepatocyte apoptosis(r = 0. 356,r = 0. 409,P < 0. 001). The positive expression rates of Caspase-3 and Caspase-8 in non-alcoholic steatohepatitis increased with the increase of pathological lesions,and the difference was statistically significant(χ2= 6. 978,χ2= 11. 043,P<0. 05). The expression of Caspase-3 and Caspase-8 in non-alcoholic steatohepatitis were not statistically significant with transaminase level(P>0. 05). Conclusion Caspase-3,Caspase-8 in nonalcoholic steatohepatitis participate in the occurrence of apoptosis; Caspase-3 and Caspase-8 in non-alcoholic steatohepatitis participate in pathogenesis.
