miR-155-5p在宫颈癌中的表达及其对宫颈癌细胞侵袭与转移的影响

摘要
目的探讨miR-155-5p在宫颈癌组织中的表达,以及其对宫颈癌细胞侵袭和迁移的影响。方法检测正常宫颈组织中miR-155-5p和宫颈癌组织的表达水平,采用荧光实时定量PCR方法检测;通过向宫颈癌细胞Hela细胞系中转染miR-155-5p mimic和inhibitor干扰内源miR-155-5p的表达,采用CCK-8法检测各组宫颈癌细胞增殖能力的变化,应用Transwell小室实验检测各组Hela细胞体外侵袭能力的差异,以及划痕实验对Hela细胞迁移能力进行评估。结果相比于正常组宫颈组织,宫颈癌组织中miR-155-5p的表达显著升高,相比于正常组差异有统计学意义(P <0.05);Hela细胞体外转染实验结果发现,降低细胞内源miR-155-5p的表达,细胞的增殖活性、侵袭以及转移能力会被抑制,而提高内源miR-155-5p的表达,细胞的增殖活性、侵袭以及转移能力会显著提高,相比于对照组差异有统计学意义(P <0.05)。结论 miR-155-5p在宫颈癌组织中高表达,其可能通过提高宫颈癌细胞的增殖活性、侵袭与迁移能力在宫颈癌的发生发展过程中起着重要的生理功能。
        Objective To investigate the expression of miR-155-5p in cervical cancer and its inìuence on the invasion and migration of cervical cancer cells. Methods The expression level of miR-155-5p in cervical cancer tissues and normal cervix tissues were detected by real-time ìuorescence quantitative real-time PCR(qRT-PCR). The transfection and expression of cervical cancer cells of Hela cell lines miR-155-5p mimic and inhibitor to realize interference of endogenous miR-155-5p, CCK-8 method was used to detect cervical cancer cell proliferation. The invasive ability of Hela cells in vitro were detected by Transwell assay, and scratch test was used to assess the cell migration. Results Compared with the normal cervical tissue, the expression of miR-155-5p in cervical cancer tissue was signiícantly increased(P < 0.05). Hela cells in vitro transfection experiments showed that to reduce expression of endogenous miR-155-5p could inhibit the proliferation activity, invasion and metastasis of cells. While to increase the expression of endogenous miR-155-5p, the proliferation activity, invasion and metastasis ability of the cells were significantly improved with statistically difference(P < 0.05). Conclusion miR-155-5p is highly expressed in cervical cancer tissues and may play an important physiological function in the development of cervical cancer by increasing the proliferation activity, invasion and migration of cervical cancer cells.
