姜黄素对喉癌细胞的KAI1/CD82表达、增殖、侵袭、转移的影响研究

摘要
目的探讨姜黄素对喉癌细胞中KAI1/CD82表达水平及对喉癌细胞增殖、侵袭、转移的影响。方法喉癌细胞培养传代后,以0、5、10、15μm/L的姜黄素作用24、48、72h,采用蛋白质免疫印迹法(Western blotting)测定KAI1/CD82的表达水平,采用四甲基偶氮唑蓝法(MTT)检测喉癌细胞的体外增殖抑制率,采用侵袭及迁移试验检测喉癌细胞的体外侵袭、转移抑制率。结果在24、48、72h时,随着姜黄素水平的增加,在相同时间内KAI1/CD82表达水平均逐渐降低,增殖抑制率、侵袭抑制率、转移抑制率均逐渐提高,差异均有统计学意义(P<0.05)。5、10、15μm/L姜黄素作用后,在相同姜黄素水平下,随着时间的推移,KAI1/CD82表达水平均逐渐降低,增殖抑制率、侵袭抑制率、转移抑制率均逐渐增高,差异均有统计学意义(P<0.05)。结论高姜黄素水平能够显著降低喉癌细胞KAI1/CD82的表达及喉癌细胞增殖、侵袭及转移能力。
Objective To investigate the effect of curcumin on the expression of KAI1/CD82 in laryngeal carcinoma cells and the proliferation,invasion and metastasis of laryngeal carcinoma cells.Methods After cultured passage of laryngeal carcinoma cells,the expression of KAI1/CD82 was determined by Western blotting with curcumin at 0,5,10,15μm/L for 24,48,and 72 h.The in vitro proliferation inhibition rate of laryngeal carcinoma cells was detected by nitroblue assay(MTT).The invasive and metastasis inhibition rates of laryngeal carcinoma cells were detected by invasion and migration assays.Results At 24,48 and 72h,with the increase of curcumin level,the expression level of KAI1/CD82 decreased gradually,and the inhibition rate of proliferation,inhibition rate of invasion and metastasis rate increased gradually,the differences were statistically significant(P<0.05).After 5,10,15μm/L level curcumin treatment,under the same curcumin level,the expression level of KAI1/CD82 gradually decreased with time,and the inhibition rate of proliferation,inhibition rate of invasion and metastasis rate increased gradually,the differences were statistically significant(P<0.05).Conclusion Curcumin can obviously restrain the expression level of KAI1/CD82,and the inhibitory of proliferation,invasion,transfer.
