一株产吲哚乙酸菌的筛选、鉴定与优化

摘要
为筛选出一株产吲哚乙酸(IAA)能力较好的菌,对采自辽宁省大连市郊区农田的植物根际土壤中的植物根际促生菌进行分离筛选,得到16株有产IAA活性的菌。从中选出产IAA量最高的菌株对其进行鉴定,并通过单因素试验与响应面法优化该菌株产IAA的培养条件。结果表明,该菌为芽孢杆菌属,最适生长条件为培养时间45 h,以0.29%的牛肉膏为碳源,1.00%酵母浸粉为氮源,添加金属离子为0.78%硫酸镁。优化后菌株IAA产量由未优化前的83.65 mg/L提高至106.11 mg/L。
        To screen a strain with good indole-3-acetic acid(IAA) producing ability, 16 strains of IAA-producing bacteria were isolated from the rhizosphere soil of plants collected from suburban farmland in Dalian of Liaoning Province. The strain with the highest IAA production was selected and identified. The cultivation conditions of IAA production were optimized by single factor test and response surface methodology. The results showed that the strain was Bacillus sp. The optimum growth conditions were as follows: culture time of 45 h, 0.29% beef extract as carbon source, 1.00% yeast extract as nitrogen source and 0.78% Mg~(2+) as metal ions. The IAA yield increased from 83.65 to 106.11 mg/L after optimization.
