摘要
目的建立1种快速筛选抗菌活性海洋真菌菌株的方法。方法采用改良双层琼脂板法,将接种细菌的培养基直接加入真菌培养基中进行共培养,并通过观察真菌菌落周围抑菌圈大小,快速筛选抗菌活性菌株。结果利用改良双层琼脂板筛选法,从一批真菌中快速筛选获得1株具有显著抗菌活性的菌株WZXY-33-10,并通过现代色谱学及波谱学方法从其发酵物中分离鉴定了2个抗菌活性化合物equisetin(1)和epi-equisetin(2)。结论改良双层琼脂板筛选法可以作为1种快速发现抗菌活性海洋真菌的方法。
Objective To establish a rapid screening method for fungal strains with antibacterial activity.Methods The double-layer agar plate screening method was used.The agar culture media that containing bacteria was added into fungal agar layer and coculture for some time,target fungal strain that possessed significant antibacterial circle would be selected out for further investigation.Results One antibacterial strain WZXY-33-10 was selected out from dozens of fungal strains by double-layer plate screening assay for the purification of antibiotics.Additionally,two antibacterial compounds equisetin(1)and epi-equisetin(2)were isolated from the fermentation extract of WZXY-33-10 by modern chromatography methods.Conclusion The double-layer plate screening can be a rapid method for screening of antibacterial marine fungal strains.
