摘要
观察小花清风藤不同药用部位及溶剂提取物对四氯化碳致小鼠急性损伤的保护作用.取昆明雄性小鼠共168只,随机分为21组,每组8只,即正常组、CCl_4模型组、水飞蓟素组、水提取组低、高剂量组、50%乙醇提取组低、高剂量组、95%乙醇提取组低、高剂量组.正常组灌胃给予等体积的0.5%羧甲基纤维素钠,其余组均按20mL/kg灌胃给予相应的药物,每日1次,连续7d,每天观察实验动物行为及体征,末次给药的次日,取材进行实验.取肝,计算肝脏的称量和质量指数,取肝组织匀浆检测肝组织中ALT、AST和SOD、GSH-Px活性及MDA含量,另取肝组织进行组织形态学分析.结果表明小花清风藤可减轻CCl_4对肝脏的脂质过氧化损伤,降低肝组织中ALT和AST活性,升高肝组织中SOD和GSH-Px水平,降低肝组织中MDA含量,从而减轻CCl_4对肝组织氧化损伤,改善肝脏的组织病理学形态,保护肝细胞并恢复肝功能.在小花清风藤水提取组低、高剂量组,50%乙醇提取组低、高剂量组,95%乙醇提取组中,以SJY作用效果比较好,依次是50%AJY、95%AJY、SJ、50%AJ、95%AJ、SY、50%AY、95%AY,且呈现明显的量效关系.小花清风藤对CCl_4致小鼠急性肝损伤具有显著的保护作用,其作用机制可能与其抗氧化作用有关.
        To observe the protective effect of different medicinal parts and solvent extracts in Sabia.Parviflora Wall.ex Roxb on carbon tetrachloride induced acute injury in mice,a total of 168 male Kunming mice were randomly divided into 21 groups,with 8 rats in each group,namely,normal group,CCl_4 model group,silymarin group,low,high dose of water extraction group groups,low,high dose of 50% ethanol extraction groups groups,low and high dose 95% ethanol extraction group groups.The normal group was given the equal volume of 0.5% carboxymethyl cellulose sodium,the other groups were given the corresponding medicine to the stomach by 20 mL/kg,once a day,continuous 7 d,we observed the behavior and signs of the experimental animals every day.The last day of the last drug delivery was carried out;liver were taken to calculate the weight and mass index of liver;and the liver tissue homogenate was used to detect ALT,AST,SOD,GSH-Px activity and MDA content;and the liver tissue was histomorphologically analyzed.The results show that the Sabia.Parviflora Wall.ex Roxb could reduce the lipid peroxidation damage of CCl_4 to liver,reduce the activity of ALT and AST in liver tissue,increase the level of SOD and GSH-Px in liver tissue,reduce the MDA content in liver tissue,reduce the oxidative damage of liver tissue,improve the histopathological form of liver,protect the liver cells and restore the liver function.In the low and high dose of SJY,SJ,SY groups,low and high dose of 50%AJY,50%AJ,50%AY groups and low and high dose of 95%AJY,95%AJ,95%AY,the effect of SJY is better,and then 50%AJY,95%AJY,SJ,50%AJ,95%AJ,SY,50%AY,95%AY.The aforementioned results show a significant dose effect relationship.Conclusion:Sabia.Parviflora Wall.ex Roxb has protective effect on acute liver injury induced by CCl_4,and its mechanism may be related to its antioxidant activity.
