摘要
目的:探讨黄芩苷对大鼠正畸牙保持阶段牙周组织改建以及成骨因子Runx2表达的影响。方法:将64只雄性Wistar大鼠(8周龄),随机分为8组,即空白组、阴性对照组和6组实验组,每组均8只大鼠。建立大鼠正畸牙移动模型21 d后,实验组于去除装置前1 d开始腹腔注射40 mg/kg黄芩苷溶液,1次/d;对照组每天注射等量生理盐水。保持1,3,7,14,21,28 d后处死,测定牙齿复发距离,HE染色观察牙周组织改建,免疫组化染色检测Runx2的表达。结果:除1 d组外,实验组复发距离均小于对照组,差异有统计学意义(P<0. 05)。在正畸保持期,对照组牙根远中侧牙周组织Runx2的表达强度呈逐渐减少的趋势,而实验组牙周组织Runx2表达强度呈逐渐增强的趋势。除1 d组外,实验组Runx2表达强度明显高于对照组,差异有统计学意义(P<0. 05)。结论:黄芩苷能够显著增强大鼠正畸牙移动保持期牙周组织中Runx2的表达,促进牙槽骨改建,从而抑制正畸牙齿的复发。
        Objective: To explore the influence of Baicalin on the expression of Runx2 in rats during the retention stage after orthodontic tooth movement. Methods: Totally 64 male Wistar rats( 8 weeks old) were divided into 8 groups randomly,each group had 8 rats that were the blank group,the control group( CG,1 group) and the experimental group( EG,6 groups). Orthodontic tooth movement model was established for 21 days. And 40 mg/kg Baicalin were peritoneal injected once every day. Same volume of saline was injected in the CG. These forces were kept for 1,3,7,14,21,28 days and followed by animal sacrifice. The recurrent distances were measured. Morphology changes of periodontal tissue were observed by hematoxylin-eosin( HE) staining. Expression of Runx2 in periodontal tissue was determined by immunohistochemistry staining. Results: The recurrent distances in each group of the EG were shorter than the CG except the 1 d group( P<0. 05). During the retention stage,the expression of Runx2 was decreasing in the distal side of the CG,while the EG increased. Compared with the EG of the same time,the expression of Runx2 in the EG were notably higher except the 1 d group( P<0. 05). Conclusion: Baicalin can increase the expression of Runx2 in the periodontal tissue of rats during the retention stage after orthodontic tooth movement,so as to promote the alveolar bone reconstruction,which will be beneficial to the inhibiton of orthodontic relapse.
