基于BODIPY两亲小分子的自组装纳米荧光探针用于选择性检测谷胱甘肽

摘要
生物硫醇,如谷胱甘肽(GSH)、半胱氨酸(Cys)、同型半胱氨酸(Hcy),在维持生物体内氧化还原平衡等方面发挥着重要的作用。其含量异常也与不同疾病的发生相关,因此对它们的选择性检测十分重要。由于生物硫醇的结构和反应活性的相似性,一般的荧光探针往往对它们会产生相同或相似的响应。在以往工作的基础上,我们在疏水性的氟硼荧(BODIPY)染料上引入长醚链亲水性基团,从而得到一侧亲水,另一侧疏水的新型两亲性BODIPY染料。该染料可在水中自组装形成纳米胶束,BODIPY荧光分子由于聚集引发猝灭。与GSH反应后,BODIPY染料的亲水性明显增强,导致胶束解聚,发光增强。在同等条件下,Cys和Hcy反应的解聚过程明显减慢,从而达到选择性检测GSH的目的。
Biological thiols, such as glutathione(GSH), cysteine(Cys), and homocysteine(Hcy), play crucial roles in maintaining biological redox balance in biological systems.On the basis of previous work, we introduced hydrophilic long ether chain groups into BODIPY to get a new amphiphile which can self-assembly form nano-micelles in water, and its fluorescence was quenched by aggregation. After reacted with GSH, nano-micelles disaggregated and fluorescence increased significantly. Under identical conditions, the disaggregation process for Cys and Hcy was much slower than GSH, thus the selective detection of GSH was achieved.
