含类异戊二烯探针分子合成及其与酶的相互作用

摘要
类异戊二烯链包括异戊烯基、香叶基、法尼基、香叶香叶基等,是C_5烯烃重复单元结构。含有此结构的化合物是萜类、甾醇类、类胡萝卜素、多萜醇类等多种天然产物分子的合成前体,在维他命K,辅酶Q,生育三烯酚类,以及黄酮类化合物中均保留有完整的结构单元,此外,也可作为蛋白质翻译后修饰的供体。设计合成含类异戊二烯基的探针分子,利用石英晶体微天平(QCM)和等温量热滴定仪(ITC)测定类异戊二烯基与五种酶的相互作用,并借助分子模拟分析类异戊二烯与酶的关键氨基酸残基相互作用机理。本研究对于阐明生物合成含有类异戊二烯基的天然产物或功能分子的酶反应机制具有重要意义,为理性设计合成生物学模块,构建含类异戊二烯基产物生物合成代谢通路提供理论指导。
Isoprenoid chains such as isopentenyl,geranyl,farnesyl,and geranylgeranyl,C_5 olefin repeat units,are building blocks for many natural molecules,terpenoids,sterols,carotenoids,and dolichols.They are present as intact tails in vitamin K,coenzyme Q,tocotrienols,and flavonoids.They also can be donor in protein post-translational modification.We designed and synthesized isoprenoid chain-contained probes,and explored interaction between probes and five enzymes using QCM and ITC.Analysis of the interaction mechanism of the enzyme key amino acid residues and probes by molecular simulation.It is important significance for elucidating enzyme reaction mechanism of biosynthesis of natural product or functional molecules containing isoprenoid.It could provide theoretical guidance for rational design of synthetic biology module,and constructing isoprenoid-contained product biosynthesis metabolic pathways.
