桑黄菌株的物理诱变及优势菌株的筛选

摘要
桑黄(Sanghuangporus sanghuang)是一种大型珍稀药用真菌,具有独特的药用价值和广泛的生物活性,富含多糖、黄酮等活性物质。为提高桑黄活性物质产量,本研究采用3种不同的物理方法对桑黄菌原生质体进行诱变,以多糖、总黄酮含量及生物活性为指标对诱变菌株进行筛选。本文将三种不同诱变方法紫外(UV)、钴60(60Co)、常压室温等离子体(ARTP)进行了比较,结果显示正诱变率ARTP>UV>60Co。通过ARTP诱变初筛获得了1139株优势菌株,五代筛选后获得了3株稳定遗传的优势菌株A130、A157、A67。与对照相比,多糖含量分别提高了32.65%、23.02%、48.54%,黄酮含量分别提高了63.19%、57.00%、34.57%。拮抗实验和RAPD分析证实了优势菌株的遗传物质发生了改变。HPLC分析从代谢产物角度证实了菌株的诱变效果。筛选获得优势菌株与对照相比,在清除自由基和抑制肿瘤细胞增殖等体外生物活性方面均表现出不同程度的提高。
