玛咖多糖提取及近红外在线检测方法构建

摘要
本文的目的是通过聚能逆流双频、聚能逆流单频和发散三频三种不同工作模式超声设备提取玛咖多糖,同时进行单因素实验来寻找最佳超声处理参数。采用近红外透射光谱法,建立玛咖提取过程多糖含量的质控指标的快速定量分析模型,并进行提取终点的快速判断,实现提取过程的在线检测。结果表明,最优的超声模式是聚能逆流双频20kHz/35kHz,在此模式下超声处理单位体积超声功率150W/L,超声时间15min,超声温度50℃,料液比1:30,多糖提取率达60.64%。采用联合偏区间最小二乘法建立多糖含量提取过程的在线监测模型,矫正集和预测集的相关系数分别为0.9621和0.9627,交互验证均方根误差和预测均方根误差分别为0.0116和0.0137。NIR光谱分析技术可作为一种多糖提取过程生产过程的在线分析方。
The objective of this study was to compare the effect of different working mode of energy aggregation counter flow single-frequency ultrasound,energy aggregation counter flow dual-frequency ultrasound and divergence triple-frequency ultrasound on the content of polysaccharide from maca(Lepidium meyenii).The one-factorial experiments were taken to search the best pretreatment parameters of ultrasound.To establish a rapid quantitative analysis model for the content of polysaccharide of Maca extraction with application of near-infrared(NIR) spectroscopy,and quickly determine the end point of extraction process,so as to realize on-line monitoring of the extraction process of Maca.The best ultrasonic working mode was20kHz/35 kHz at energy aggregation counter flow dual-frequency ultrasound.With this working mode,the extraction polysaccharide content could increase to 60.64%under the following optimal ultrasound pretreatment conditions:ultrasonic power per unit volume of 150W/L,ultrasound time of 15 min,ultrasonic temperature at 50℃,the ratio of solid to liquid 1:30.Diels for an example,synergy interval partial least squares(Si-PLS) models were developed for the content of polysaccharide,the correlation coefficients of the calibration and prediction models was achieved with 0.9621 and 0.9627,with root mean squared error of cros-validation(RMSECV) and prediction(RMSEP) was 0.0116 and 0.0137,respectively.The study demonstrates that NIR spectroscopy can be used s an online analytical method to determine the content of polysaccharide content in maca.
