不同倍性萱草杂交及胚培养研究

摘要
萱草属植物,观赏价值高,使用范围广泛。但不同倍性萱草间杂交结实率低,在许多植物中,幼胚离体培养技术是克服受精后生长障碍的常用方法。试验以材料间常规花期杂交平均结实率低的二倍体、三倍体、四倍体萱草品种为亲本,设置不同倍性间的杂交组合,开展了从杂交育种到胚培养等方面研究,得到结果如下:萱草不同杂交组合的亲和性有一定差异,杂交中会出现4种情况:不能完成受精;仅卵细胞受精,只产生胚而无胚乳;仅极核受精,只产生胚乳而无胚;完成双受精,产生胚和胚乳。杂交败育的原因之一是双受精的比率较低,胚胎正常发育的情况较少,导致胚无法得到胚乳提供的营养,抑制了胚的正常发育。胚培养试验确定了最适胚珠离体培养时间是授粉后7d;配方为MS+0.01mg/L NAA+0.032mg/L 6-BA+蔗糖30g/L的培养基效果优于其他,因而通过胚培养可以有效降低杂交的败育率,提高杂种胚的成活率。
Hemerocallis species have high ornamental values as well as wide range of uses,but the hybridization of daylily shows a high rate of abortion.Many plants' studies suggested that embryo culture is a proper method which can provide a guarantee of effective access to hybrid progeny.The cross combinations including diploid,triploid,tetraploid daylilies are used as parents in hybridization and embryo culture.Four conditions were found during the development.The first condition was that fertilization could not be completed.The second one was that only the egg cell was fertilized.The third one was that polar nuclear cells were fertilized and endosperm was obtained.The last condition was that double fertilization was completed and both embryo and endosperm were obtained.The possible cause of hybridization abortion is the low rates of double fertilization,which may result in the failure of endosperm providing nutrients and finally inhibiting the development of normal embryos.It result showed that the best time to rescue the hybrid embryos is 7~(th) day after cross-breeding.The formula of medium which contained MS +0.01mg/L NAA +0.032mg/L 6-BA + sucrose 30g/L for embryo culture had better effects.Therefore,the abortion rates of hybridization of daylilies can be decreased by embryo culture and the percentages of hybrids can be improved as well.
