利用斑马鱼研究红色荧光蛋白标记的副溶血弧菌和嗜中性粒细胞之间的交互作用

摘要
副溶血弧菌(Vibrio parahaemolyticus,Vp)是一种重要的水生动物来源的人畜共患的病原菌,能引起海水动物及人类严重的弧菌病。为了研究嗜中性粒细胞在Vp感染中的作用,我们将红色荧光蛋白(RFP)标记了一株致病菌Vp57,利用斑马鱼模型测试标记前后毒力变化及引起的疾病症状,并观察嗜中性粒细胞与Vp感染之间的动态过程。结果显示细菌毒力在RFP标记前后没有变化,并能在宿主中引起相似的症状。用显微注射的方法感染3dpf的斑马鱼尾静脉,当注射579cfu/个体时,Vp57~(RFP)通过血液循环进入到全身,注射1h后嗜中性粒细胞在感染部位增多,并在血液循环中清除部分细菌,3h后鱼死亡,体节处发现少量Vp57~(RFP);当注射974cfu/个体时,2h后体节处也发现大量Vp57~(RFP),2.5h后脊椎严重弯曲并死亡,推测脊椎弯曲和细菌毒性相关,嗜中性粒细胞的清除作用在感染后3h最明显。结果表明斑马鱼是一个很好的研究先天免疫中嗜中性粒细胞和Vp57交互作用的模型。
Vibrio parahaemolyticus is an aquatic zoonotic pathogen causingvibriosis in marine animals and human.To elucidate the function of neutrophils ininnate immune response, we tagged a pathogenic strain Vp57 with RFP, observed their dynamic interaction with the neutrophils in3 dpfzebrafishlarva. Two strains showed similar LD50 and induced similar histopathology results in hosts. We microinjected 579 cfu Vp57~(RFP) into zebrafish, Vp57~(RFP) distributed in the whole blood vessels as blood circulating, neutrophils fought with bacteria at 1h post infection(hpf), fish diedafter 3h. When we injected 974 cfu, somites of larvae were covered with bacteriaat 2hpf.After2.5hpf, fish dead and had serious spine curvature,this maybe related to bacterial virulence.Scavenging effect of neutrophils was obvious at 3hpf.The results showed that zebrafish model is useful for studying neutrophils function in innate immune response to Vp infection.
