调控喉鳞状细胞癌Livin基因在裸鼠移植瘤表达的实验研究

摘要
目的:利用反义Livin多核苷酸调低喉鳞状细胞癌裸鼠移植瘤Livin基因的表达以探讨其对喉鳞状细胞癌凋亡的影响。方法:(1)雌性裸鼠(BALB/c-nu/nu),分成Livin AS-DNs(Livin反义多核苷酸)、Mis-DNs、Sen-DNs、对照组,每组7只,于裸鼠背部皮下注射1x10~(4-5)个hep-2细胞成瘤;(2)采用Western blotting检查AS-DNs、Mis-DNs组、Sen-DNs组、对照组瘤组织Livin蛋白;(3)利用TUNEL染色法对各组瘤体中细胞凋亡情况进行分析。结果:(1)Livin蛋白检查量最低为AS组(15.02±1.68)%,Sen组、Mis组、Control组明显高于AS组,AS组与其他组比,P<0.05,有统计学意义;(2)裸鼠瘤体内喉鳞癌细胞的凋亡指数最高为(20.15±3.1)%,为AS-DNs组,Sen组、Mis组、Control组明显低于AS组,AS组与其他组比,P<0.05,差异有统计学意义;(3)采用AS-DNs治疗组的肿瘤生长较Sen组、Mis组、Control组慢,AS组与其他组比,P<0.05,差异有统计学意义;结论:反义Livin多核苷酸在裸鼠移植瘤内促进了喉鳞状细胞癌的凋亡,抑制了肿瘤的生长。
Objective: To investigate the effect of apoptosis of the antisense polynucleotide( AS-DNs) targeting Livin to transplantation tumor of laryngeal squamous cell carcinoma cell line( Hep-2) in nude mice. Methods:(1) Female BALB/c-nu/nu mice were divided into four groups( n= 7 in each group):(a) Control group,(b) Livin AS-DNs group,(c) Sen-DNs group, and(d) Mis-DNs group. About 1 ×10~(4-5) Hep-2 cells were subcutaneously inoculated in dorsal space of the mice.(2)The expression of Livin protein in tumor was examined by Western blotting.(3) TUNEL assay was used to analyze cell apoptosis in tumor of nude mice. Results:(1) The Livin protein expression of AS-DNs was(15.02 ± 1.68)% to the untreated group,which was much lower than other group. There are statistical significance in the group AS-DNs with Control group、Sen-DNs group, and Mis-DNs group(P<0.05).(2) The apoptotic cells was notably higher in AS-DNs group than that of other group. The AI(Apoptotic Index) was(20.15 ± 3.1)% in AS-DNs group,which was significantly higher than that of the other groups.(3)Tumor growth rate was slower in AS-DNs group than that of Control group、Sen.-DNs group, and Mis-DNs group. Conclusion: AS-DNs of Livin could promote apoptosis of laryngeal squamous cell carcinoma and inhibit tumor growth in vivo.
