UHPLC-QTOF/MS的茶叶籽酚类化合物定性研究

摘要
茶叶籽油具有很好的抗氧化性,近年来得到越来越多的关注。一般认为其中的多酚类物质对茶叶籽油抗氧化性起着主导作用。本工作采用超高效液相色谱-四级杆飞行时间串联质谱(UHPLC-QTOF/MS)技术定性分析茶叶籽中酚类化合物。茶叶籽样品与正己烷摇匀后,采用60%甲醇溶液提取。提取液经旋转蒸发干燥后,重溶到50%甲醇水溶液用于UHPLC-QTOF/MS分析。实验选用Spursil-C_(18)(150mm×2.1mm×2.1μm)色谱柱和0.1%甲酸水与0.1%甲酸乙腈的二元流动相进行梯度洗脱,并在负离子模式下进行飞行时间质谱鉴定。基于QTOF高分辨特性及精确质量定性特点,并结合文献和碎片离子分析,初步推断出茶叶籽甲醇提取物中的23种酚类化合物,其中包括13种酚酸化合物、4种儿茶素类化合物和6种黄酮类化合物。进一步通过与标准品的保留时间比对,确证了这些化合物。这些结果表明,高质量精度及高分辨的UHPLC-QTOF/MS技术,可以为鉴定茶叶籽中酚类化合物组成提供一种准确、可靠的定性分析方法。
Tea seeds oil is reported to have anti-oxidative effect,and has drawn lots of attention recently.It is thought that polyphenols in the tea seeds play the major role in the anti-oxidative function.In this work,a UHPLC combined with QTOF/MS instrument(UHPLC-QTOF/MS) was applied to qualitatively determine the phenolic compounds of Camellia sinensis seed.The seed sample was extracted and subjected to UHPLC-QTOF/MS analysis under gradient elution.The eluent was directed to QTOF/MS under negative ion mode for compound identification.A total of 23 phenolic compounds were tentatively identified,including 13 phenolic acids,4 catechins and 6flavonoids,which were characteristic in the methanol extract,based on the references and relevant structural information of molecular and fragment ions.These compounds were further confirmed by matching against the standard compounds in retention times.The results demonstrated that high mass accuracy and high resolution UHPLC-QTOF/MS is a reliable technology for qualitative identification of the phenolic compounds in C.sinensis seed.
