内部萃取电喷雾电离质谱直接分析肺组织样品化学组分

摘要
离子化技术是近年来常压快速质谱分析技术的重点,基于不同离子化技术建立了许多各具特色的质谱分析方法。然而,直接分析样品内部化学组分的电离技术鲜有报道。内部萃取电喷雾电离(iEESI)~([1])能够直接对组织样品内部的化学物质进行萃取和离子化,结合质谱高灵敏度、高特异性的特点,实现定性和定量分析。本文以肺组织样品中的氨基酸为研究对象,在无需样品预处理的情况下使用iEESI-MS进行内部分析,并使用DESI-MS~([2])进行表面分析对比实验。实验结果表明,iEESI-MS能够快速、灵敏地检测到多种氨基酸,证明了该方法在全面获取组织样品信息方面的优势。另外,利用iEESI-MS对肺癌和癌旁组织进行分析,并结合主成分分析(PCA)快速、准确地区分了肺癌和癌旁组织。以上结果表明,iEESI-MS是研究生物组织内部成分的理想工具,并有望应用于临床诊断、生物学、代谢组学等领域。
iEESI-MS is characterized by extraction and ionization processes occur simultaneously,and can directly qualitative and quantitative analysis the interior chemicals of bulk sample,combining mass spectrometry with high sensitivity and high specificity.In this paper,bulk samples of lung tissue with amino acid were analyzed by iEESI-MS.Moreover,iEESI-MS is compared to DESI-MS proved that it can detect variety of amino acids of lung tissue quickly and sensitively.In the PCA score plot,lung cancer samples were clearly separate from the adjacent normal tissues that were analyzed by iEESI-MS.These findings suggest that it provides a powerful tool for studying internal chemicals of biological tissues,and offer possibilities applications in the field of metabolomics for cancer,medical engineering,and clinical diagnosis.
