快速检测天然物中紫杉醇类活性物质的方法

摘要
本文利用碳纤维/活性碳纤维柱多维组分分离系统,建立了一种对天然物中痕量紫杉醇类物质快速在线分离的方法,并与串联质谱联用实现对多种类型的紫杉醇类活性物质的快速检测。样品经甲醇超声萃取后直接进多维组分分离系统,以7种代表性的紫衫烷物质作的模板分子进行研究,该方法将极性差异较大的10-脱乙酰基巴卡亭Ⅲ、巴卡亭Ⅲ和7-木糖基-10脱乙酰基紫杉醇、7-木糖基-紫杉醇、三尖杉宁碱、紫杉醇及7-表-10-脱乙酰基紫杉醇初步分离,同时去除弱极性的脂质、色素等干扰物;含目标物的组分直接进串联质谱进行定性、定量分析。结果表明:该方法能够快速准确分析复杂植物样品中的紫杉醇类物质,7种目标物的松针加标回收率为75%~118%,RSD为14%~20%(n=6),LOD为5 ng mL~(-1)~15 ng mL~(-1)。此方法只需6分钟即可从复杂的样品中分离检测多种类型的紫杉醇类物质,是一种快速、高效、重现性好、有机溶剂用量少(40μL)且操作方便、绿色环保的方法。
We developed a fast and online-separation method for detection multiple trace taxanes in natural products extracts based on CF/ACF column multidimensional fractionation system.Methanol extracts were directly into the system for separation according to polarity.Targets were preliminary separated while removing weakly polar lipids,pigments ect.Then targets were into the MS/MS for qualitative and quantitative analysis.The recoveries of seven targets(typical taxanes) in pine needle spiked samples are 75%~ 118%,RSD are 14%~ 20%(n = 6),LOD are 5ng mL~(-1) ~15 ng mL~(-1).This method only needs 6 min for detection multiple taxenes in complex samples.It is a fast,efficient and green approach with less organic solvent(40 μL).
