精氨酸水平对团头鲂幼鱼胰岛素信号通路相关基因的影响

摘要
为探讨精氨酸水平对团头鲂幼鱼生长、胰岛素信号通路相关基因的影响,选取相同规格的团头鲂(20.0±0.03g)分至18个网箱(1 m×1 m×1 m),每个网箱20条。分别投喂精氨酸梯度0.87%到2.70%.的6组饲料,养殖期8周。结果表明,团头鲂终体重(FW),蛋白效率(PER)、增重率(WG)、特定增长率(SGR)提高,均在1.62%组达到最大,饵料系数(FCR)呈相反的趋势。根据FCR和SGR,利用二次回归曲线分析,最适精氨酸水平分别为1.85%(占饲料蛋白5.60%)和1.84%(占饲料蛋白5.58%)。饲料精氨酸水平影响胰岛素受体底物1(IRS-1)和雷帕霉素靶蛋白(TOR)的表达,并与生长性能趋势相似。在1.62和1.96%组,胰岛素样生长因子1(IGF-1)表达上调。在2.31%组,磷脂酰肌醇3-激酶(PI3K)表达上调。在过量的精氨酸水平(2.70%),核糖体S6激酶(S6K1)呈现高表达现象,与IRS-1和TOR呈相反趋势。结果表明,最适组(1.62%)提高团头鲂幼鱼的生长并且上调TOR信号通路的表达。然而,过量的精氨酸水平(2.70%),会导致S6K1的高表达,通过负反馈调节而形成胰岛素抵抗,包括减少IRS-1以及PI3K相关基因的表达。
An 8-week feeding trial was conducted to investigate the effects of dietary arginine levels on the growth performanceand insulin pathwaysignalingmolecules in juvenile blunt snout bream.Fish(initial weight 20.0 ± 0.03g) were randomly sorted into 18 cages(1 m × 1m × 1 m) and each cage was stocked with 20 fish for farm pond culture.Arginine levels ranging from 0.87% to 2.70% of dry diet. Results showed that final weight(FW), weight gain(WG), specific growth rate(SGR) and protein efficiency ratio(PER) increased with increasing dietary arginine level up to 1.62%, and thereafter showed a decreasing trend, while feed conversion ratio(FCR) showed a converse trend.Based on FCR and SGR, the optimal dietary arginine level was determined to be 1.85% of diet(5.60% of dietary protein) and 1.84% of diet(5.58% of dietary protein), respectively, using quadratic regression analysis.Expressions of insulin receptor substrate 1(IRS-1) and target of rapamycin(TOR) mRNA were regulated by dietary arginine levels.
