酸性电解水对大肠杆菌生物被膜的清除作用研究

摘要
大肠杆菌是重要的食源性致病菌,易形成生物被膜(Biofilm)从而给食品安全带来严重风险。本文采用荧光显微镜、扫描电镜和涂布法研究了不同NaCl浓度(0.01%,0.10%,0.30%,0.50%)的酸性电解水(acidic electrolyzed water,AEW)和不同处理时间(2,5,10min)对大肠杆菌K12生物被膜的清除效果。结果表明,随着NaCl浓度的增高,生物被膜内的活菌数逐渐减少,分别为6.37,5.35,3.99,3.75(1g(CFU/ml))。用0.3%NaCl的AEW处理2 min,5 min,10min后活菌数逐渐减少,处理5min和10min没有显著性差异(p>0.05)。0.30%NaCl的AEW作用5min后活菌数显著降低(p<0.05)。此外,荧光显微镜和扫描电镜结果显示,该条件下处理后的生物被膜结构明显变稀疏,厚度减少,并且细胞发生裂解。本研究表明AEW不仅可以有效的去除生物被膜而且还能破坏细胞膜,降低细胞活性。因此,AEW可以作为一种新型高效杀菌剂,为食品安全提供高效的安全保障,在食品加工业中具有潜在的应用前景。
Escherichia coli is an important foodborne pathogens,which can form biofilm and bring serious risks to food safety.The purpose of this study was to evaluate the efficacies of acidic electrolyzed water(AEW) produced by different NaCl concentration(0.01%,0.10%,0.30%,0.50%) and exposure time(2,5,10 min) in biofilm of Escherichia coli strain K12,using plate count method,fluorescence microscope and scanning electron microscopy on polystyrene microliter plates.The results showed that with the increasing of NaCl concentration(0.01%-0.50%),the population of E.coli reduced significantly(p <0.05),and the viable counts were 6.37,5.35,3.99 and 3.75(lg(CFU/ml)),respectively.There was no significant difference of populations in biofilms treated by 0.30%NaCl AEW for 5min and 10 min(p >0.05).Besides,visualizing morphological changes of bacterial cells by scanning electron microscopy and fluorescence microscope after treated with AEW,the cells were detached from the biofilm matrix and the integrity of cell envelopes were decreased.Taken together,AEW not only can effectively remove biofilm,but also can destroy the cell membrane integrity to reduce the cell activity.Therefore,the application of treatments that last a few minutes using AEW represented an promising disinfection strategy against pathogens associated with medical and industrial settings.
