以白蛋白为载体纳米探针在肿瘤生物影像与治疗中的应用

摘要
白蛋白是血清的主要成分之一,因其天然的两亲性结构,成本低,来源广泛,易于加工和良好的生物相容性,被广泛用于药物载体研究。我们利用化疗药物紫杉醇诱导蛋白分子自组装的技术,构建具有肿瘤靶向能力、多模态成像和联合治疗功能的蛋白纳米药物,在针对小鼠原发乳腺癌和转移乳腺癌的光热/光动力/化疗联合治疗中体现出很好的疗效和低毒副作用。肿瘤的微环境,如高孔隙流体压力、乏氧及低pH值通常与控制肿瘤的生长和转移相关因素密切相关。肿瘤的弱酸性不仅会增加癌细胞的转移,还会导致耐药性(MDR)。因此,我们又构建了p H响应的白蛋白-染料的纳米探针不仅可以实现肿瘤p H值的实时成像,还能实现p H响应的的光热治疗。
Albumin, the most abundant plasma protein, has been extensively explored as a versatile nanocarrier for drug delivery due to its complete biocompatibility, lack of toxicity and immunogenicity, easy and inexpensive production, as well as great storage and in vivo stabilities. Albumin, in addition to its applications as conventional drug carriers, can also serve as the building block in the construction of multifunctional theranostic nano-agents, showing promises in cancer-targeted, multimodal imaging-guided, combination therapy of cancer. In addition to applications in cancer therapy, albumin-based nano-complexes have also shown great potential for biomedical imaging. A pH-responsive albumin-based nano-probe is developed to realize quantitative photoacoustic p H imaging in vivo in the tumor microenvironment.
