人参多糖体外抗猪生殖与呼吸综合征病毒活性的研究

猪繁殖与呼吸综合症(Porcine reproductive and respiratory syndrome,PRRS)是严重危害养猪业的传染病之一,也是重点防控的疾病之一。人参多糖具有抗病毒及增强免疫的作用。为了寻找有效预防和治疗PPRS的药物,本文以marc-145细胞为模型,采用细胞病变抑制试验和MTT法测定细胞活力,观察人参多糖(Ginseng polysaccharides complex,GPS)在体外抗猪繁殖与呼吸综合征病毒(Porcine reproductive and respiratory syndrome virus,PRRSV)的作用,并通过改变药物作用方式初步探讨GPS的抗病毒机制。试验分为6组,分别为50μg/mL、100μg/mL、200μg/mL和400μg/mL人参多糖组、病毒对照组及正常细胞对照组。在GPS对PRRSV感染marc-145细胞的阻断和抑制作用中,各浓度人参多糖均显著提高感染PRRSV后marc-145细胞的存活率(P<0.05),人GPS处理组marc-145细胞中的PRRSV量均显著低于病毒对照组(P<0.05)。而且,GPS可以促进marc-145细胞中IL-6和IFN-γ的mRNA表达,也能增强细胞上清中IL-6和IFN-γ的活性。本实验研究表明GPS在体外可有效阻断或抑制PRRSV对marc-145细胞的感染,提示GPS可作为预防及治疗PRRS的候选药物。
Porcine reproductive and respiratory syndrome(PRRS) is one of key epidemic diseases which is seriously harmful to pig industry.Ginseng polysaccharides complex(GPS) is believed to be of functions of anti-virus and immune improvement.In order to search for special medicinal herb that could be effective in preventing and treating PRRS,this research used marc-145 cells as testing models throμgh cytopathic effect inhibition assay and cell vitality determined by MTT method to evaluate the inhibitory effect of GPS on PRRSV propagation in vitro.The tests were divided into 6 groups:50μg/mL,100μg/mL,200μg/mL and 400μg/mL GPS treatment groups,virus control group and cell control group.The results showed that cell survival rate and the inhibitory rate increased significantly in the marc-145 cells treated with dose-dependent GPS,and the amount of PRRSV in infected marc-145 cells that treated with different dose of GPS was also decreased observably compared with virus control group.Further,GPS promoted the mRNA expression of IL-6 and IFN-γ in marc-145 cell line,and increased the IL-6 and IFN-γ level in supernatant.It demonstrated that GPS had significant antiviral effect in vitro,and it could bea potential drug in prevention and treatment of PRRS.
