基于16SrRNA基因部分序列的8种虾虎鱼科鱼类系统发育关系分析

摘要
本研究对采自海陵岛的4种虾虎鱼科鱼类进行16SrRNA基因特异扩增测序,并与从GenBank下载的11条同源序列结合,共15条序列进行序列联合分析和系统发育研究。研究表明:15条序列T、C、A、G碱基含量分别为21.5%,25.0%,30.6%,22.9%,有保守位点333个,变异位点153个,简约信息位点131个,单一突变位点22个。运用MAGA6计算8种虾虎鱼科鱼类之间的遗传距离,结果显示8种虾虎鱼科鱼类的平均遗传距离为0.1329,其中犬牙细棘虾虎鱼和红狼牙虾虎鱼的种间遗传距离最大,青弹涂鱼和大弹涂鱼的遗传距离最小。利用最小进化法和邻位连接法构建分子系统发育树,两种方法构建的系统发育树的拓扑结构完全相同,其中斑纹舌虾虎鱼位于进化树的基部,拟矛尾虾虎鱼位于进化树的顶端。研究旨为虾虎鱼科鱼类的分类提供依据,并为其进一步的研究和保护提供参考。
16SrRNA gene from 4 species of Gobiidae in the Hailing Island were amplified and sequenced, and base ratio and genetic distances were analyzed combining with the 11 homologous sequences downloaded from GenBank. The results indicated that the average contents of T, C, A and G were 21.5%, 25.0%, 30.6% and 22.9%,and there are 333 conserved sites, 153 variable sites,131 parsimony informative polymorphic sites and 22 Single mutation sites. The calculation of genetic distance between 8 species of Gobiidae by MAGA6, the results show that the average genetic distance within-species is 0.1329, genetic distance between Acentrogobiuscaninusand Odontamblyopusrubicundus is largest and between Scartelaoshistiophorus and Boleophthalmuspectinirostris is smallest.According to the Neighbour-joining and Minimum-Evolution trees,topology of the phylogenetic tree constructed by the two methods is identical. Glossogobiusolivaceus located at the buttom of the phylogeny and Parachaeturichthyspolynema located at the top of the tree.Thisstudy will be a useful resource for the classification of Gobiidae and provide reference for further research and protection.
